14-3-3蛋白家族在卵巢子宫内膜异位囊肿中的表达及意义

【摘要】 目的:研究14-3-3蛋白各亚型mRNA及蛋白在卵巢子宫内膜异位囊肿中的表达,探讨14-3-3蛋白与卵巢子宫内膜异位囊肿发生发展的关系。方法:取经病理组织学确诊卵巢子宫内膜异位囊肿患者的异位内膜与正常子宫内膜标本各30例,采用逆转录聚合酶链反应(RT-PCR)检测组织中14-3-3蛋白各亚型mRNA表达水平,对有表达差异的亚型通过Western-blotting进一步鉴定其蛋白表达水平。结果:14-3-3蛋白各亚型在卵巢子宫内膜异位囊肿异位子宫内膜组织和正常子宫内膜组织均有表达,且卵巢子宫内膜异位囊肿异位子宫内膜组织中14-3-3σ亚型mRNA表达明显下调(P<0.05);14-3-3ε、ζ两种亚型mRNA表达明显上调(P<0.01);Western-blotting进一步检测显示14-3-3σ亚型蛋白表达也明显下调(P<0.01);14-3-3ε、ζ两种亚型蛋白表达也明显上调(P<0.01)。结论:(1)14-3-3蛋白七种亚型在卵巢子宫内膜异位囊肿异位子宫内膜组织和正常子宫内膜组织中均有表达。(2)相对正常对照组织,卵巢子宫内膜异位囊肿组织中的14-3-3σ的mRNA和蛋白表达水平均降低,14-3-3ε和14-3-3ζ的mRNA和蛋白表达均明显升高。我们的结果表明14-3-3σ、ε、ζ三种蛋白亚型均可能与卵巢子宫内膜异位囊肿的发生发展相关。

【Abstract】 Objective: To investigate the role(s) of14-3-3proteins isoforms played in thedevelopment of endometriotic cysts of ovary, we quantified the expression levels of14-3-3mRNAs and proteins in endometriotic cysts of ovary.Methods: To selecte30histopathologically diagnosed patients with endometrioticcysts of ovary and30controls, then examined the mRNA expressions of all seven14-3-3isoforms in the affected or normal endometriums by RT-PCR. For thosedifferentially expressed isoforms, their protein expressions were further determinedby Western blotting.Results: All of the seven14-3-3isoforms mRNA were detected by RT-PCR both inthe endometriotic cysts of ovary ectopic endometrium and normal endometrium.However, the mRNA expression of14-3-3σ was significantly decreased inendometriotic cysts of ovary ectopic endometrial tissues when compared with thenormal endometrial tissues (P<0.05); the mRNA expressions of14-3-3ε and14-3-3ζwere significantly increased in endometriotic cysts of ovary ectopic endometrialtissues when compared with the normal endometrial tissues (P<0.01); the proteinexpression of14-3-3σ was also significantly decreased in endometriotic cysts ofovary ectopic endometrial tissues (P<0.01); the protein expression levels of14-3-3εand14-3-3ζ were also significantly increased in chocolate of ovary ectopicendometrial tissues (P<0.01).Conclusions:(1) There was a basal mRNA expressions of all the seven14-3-3isoforms both in endometriotic cysts of ovary ectopic endometrium and normalendometrium.(2) The mRNA and protein expressions of14-3-3σ decreased inendometriotic cysts of ovary ectopic endometrial tissues when compared with normalcontrol tissues, the mRNA and protein expressions of14-3-3ε and14-3-3ζ weresignificantly increased. Our results suggested that14-3-3σ, ε, ζ three isoforms of14-3-3proteins may be involved in the development of the endometriotic cysts ofovary.
