柚果实生长过程中内源激素含量变化的研究

【摘要】 本研究以福建省漳州市平和县3个蜜柚品种(琯溪蜜柚、红肉蜜柚、红绵蜜柚)为研究对象,对柚果实发育动态以及不同部位激素的变化进行研究,并探讨内源激素与汁胞粒化的关系,以期探明内源激素对汁胞粒化的影响机制。主要研究结果如下:1、在果实发育过程中,红肉蜜柚的汁胞粒化率显著高于琯溪蜜柚和红绵蜜柚,且出现的时间也较早,花后129天的粒化率为15.38%;琯溪蜜柚在开花后139d出现粒化,粒化率为26.67%;红绵蜜柚在花后149d出现粒化,粒化率为33.33%。2、建立了平和蜜柚果实三个部位(果肉、海绵层、囊衣)内源激素的高效液相色谱研究方法,结果表明,用C18色谱柱(250x4.6mm,5μm);A相为0.001mol/L(pH3.5)的磷酸缓冲液、C相为色谱甲醇、D相为色谱乙腈溶液作为流动相,IAA、ABA、GA3、tZR的检测波长分别为226nm、260nm、207nm、260nm;流速1mL/min;柱温为35℃;进样量为10μL分别进行梯度洗脱,用配有紫外检测器、四元泵、柱温箱、自动进样器等的L-2000型高效液相色谱仪(日本日立公司),进行检测,取得良好的结果。3、揭示3个品种柚果实生长发育过程中各部位内源激素的动态变化规律。在果肉中,3品种果实生长发育期间ABA含量变化趋势相似,红绵蜜柚ABA含量整体显著高于红肉蜜柚。琯溪蜜柚果实发育期间果肉中tZR含量大部分时期高于红肉蜜柚和红绵蜜柚;在囊衣中,三品种果实生长发育期间IAA含量变化差异较大,琯溪蜜柚囊衣IAA在花后90d为0.3260μg/gFW,然后逐渐下降,随着果实的成熟,至花后169d又有升高趋势,琯溪蜜柚从花后169d持续到花后179d,仅持续10d左右。红绵蜜柚tZR整体含量大于其它品种;在海绵层中,3品种生长发育期间GA3变化趋势大致相同,花后50d-90d红肉蜜柚和红绵蜜柚的含量低于其他两个品种,但花后90d以后,情况则相反,琯溪蜜柚GA3则低于红肉蜜柚和红绵蜜柚,整体上琯溪蜜柚海绵层ABA含量显著低于其它品种。4、研究发现,自花后129d开始,3个品种中远离中柱汁胞的IAA含量显著大于靠近中柱的汁胞IAA含量,ABA则相反,远离中柱的汁胞ABA含量显著小于靠近中柱的汁胞ABA含量,但是在起初的花后129d时表现不显著,随着果实发育的逐渐成熟,才表现出上述现象。自花后129d起,粒化现象逐渐严重,但几个品种tZR在果肉中表现的变化差异并不是很显著,汁胞中GA3靠近中柱与远离中柱含量水平不一致,汁胞粒化启动前期GA3含量上升加快,随后又下降,至果实采收前期,远离中柱的汁胞中GA3含量下降,靠近中柱的又有轻微的上升趋势,GA3含量的上升与粒化的严重程度有关,尤其是琯溪蜜柚和红肉蜜柚表现更为显著。
【Abstract】 In this study three kinds of pomelo (Guanxi-miyou, Hongrou-miyou andHongmian-miyou) in Pinghe County, Zhangzhou City, Fujian Province as the researchobject, study on the development dynamic and different parts of the hormone changesin the pomelo fruit, and to explore the relationship between endogenous hormone andjuice sac granulation, expect to prove Influence mechanism of endogenous hormones onjuice sac granulation. The main results were as follows:1. During fruit development, the juice sac granulation rate of Hongrou-miyouwas significantly higher than Guanxi-miyou and Hongmian-miyou, and appeared muchearlier, the granulation rate was15.38%after flowering129d, the granulation ofGuanxi-miyou appeared after flowering139d, granulation rate was26.67%,Hongmian-miyou after flowering149d have granulation phenomenon, granulation ratewas33.33%.2. Build the research methods about the endogenous hormones in three parts (pulp,sponge layer, capsule clothing) of pomelo fruit by high performance liquidchromatography, the results showed that with a C18column (250x4.6mm,5μm); therewere three of solution as mobile phases, A-phase was0.001mol/L PBS buffer(pH3.5),C-phase was chromatography methanol, D-phase was chromatography acetonitrile. Thedetection wave-length of IAA, ABA, GA3, tZR was226nm,260nm,207nm and260nm,the flow rate was1mL/min, column temperature was35°C, injection volume was10ulby gradient elution.Good results of detection was achieved by the L-2000typehigh-performance liquid chromatograph (Hitachi, Japan) with UV detector, quaternarypump, column oven, auto-sampler and so on.3. Revealed the dynamic changes of endogenous hormones during growth anddevelopment in various parts of three varieties of pomelo fruit. In the flesh, during thegrowth and development of three varieties of fruit, the content changes of ABA hadsimilar trend, as a whole the ABA content of Hongmian-miyou was significantly higherthan Hongrou-miyou. During fruit development period tZR in flesh Guanxi-miyou content mostly higher than Hongrou-miyou and Hongmian-miyou. In the capsuleclothing, during growth and development period, IAA content changes differences inthree varieties of fruit were quitely different, IAA in Guanxi-miyou capsule clothingafter flowering90d was0.3260μg/gFW, and then gradually decreased, to afterflowering169d had increased trend as ripening, Guanxi-miyou lasted only about10dfrom after flowering169d to after flowering179d. The content of tZR inHongmian-miyou capsule clothing overall was greater than the other varieties. Insponge layer of three varieties, during the period of growth and development GA3content trended roughly the same, after flowering50d to90d Hongrou pomelo andHongmian pomelo content lower than Guanxi-miyou, but after90days, the situationwas the opposite, Overall Guanxi-miyou sponge layer ABA content was significantlylower than the other two varieties.4. The study found that start from the129d after flowering, IAA content in thejuice sac of three varieties away from the column was significantly greater than near thecolumn, in the juice sac ABA was the opposite, away from the column the juice sacABA content was significantly smaller than the other part, but at first, from the129dafter flowering performance was not significant, with the gradual maturity of the fruitdevelopment, to demonstrate the phenomenon. From129d after flowering granulationphenomenon was gradually serious, but the differences of tZR among several varietiesws not very significant, the GA3levels of the juice sac near and away from the columnwas inconsistent, the GA3content early raised faster when juice sac started to granulatein the early stage, and subsequently decreased. To pre-harvest fruit, GA3juice sac awayfrom the column decreased, near the column there were slight upward trend,as severityof granulation increased in pommelo, GA3content increased,especially performance ofGuanxi-miyou and Hongrou-miyou were more significant.
