前列通窍胶囊含药血清对家兔离体膀胱肌作用的实验研究

【摘要】 目的:运用血清药理学方法,观察前列通窍胶囊含药血清对家兔离体膀胱逼尿肌及括约肌活动的影响,并初步探讨其作用机制。方法:制备前列通窍胶囊含药血清,将家兔离体膀胱逼尿肌及膀胱括约肌(即尿道内括约肌)分别置于恒温平滑肌槽中,运用Powerlab/8s生物信号采集处理系统记录肌条的活动,观察前列通窍胶囊含药血清对各标本的影响,以及加入不同阻滞剂后含药血清对各标本的作用,分析作用机制。结果:1、连续给家兔服用前列通窍胶囊3天、5天、7天后制备的含药血清(以下简称含药血清3天组、5天组、7天组)均可增强其离体膀胱逼尿肌的收缩幅度、收缩频率及活动力,降低膀胱括约肌张力;与空白对照组、空白血清组相比,含药血清7天组的作用差异具有统计学意义(P <0.05)。含药血清3天组与7天组的作用差异具有统计学意义(P <0.05)。2、与空白血清组相比,低、中、高浓度含药血清7天组(以下简称低、中、高浓度组)均可增强膀胱逼尿肌的收缩幅度、收缩频率及活动力,降低膀胱括约肌张力;与空白含药血清相比,中、高浓度组对膀胱肌作用的差异有统计学意义(P <0.05)。与空白血清组相比,相同浓度前列通窍胶囊含药血清组及癃闭舒胶囊含药血清组均可增强膀胱逼尿肌收缩幅度、收缩频率及活动力,降低膀胱括约肌张力,差异有统计学意义(P <0.05);二者作用差异无统计学意义(P>0.05)。3、与中浓度含药血清组相比,维拉帕米可以阻断前列通窍胶囊含药血清对膀胱逼尿肌的收缩作用,差异有统计学意义(P<0.05);前列通窍胶囊含药血清可以降低去甲肾上腺素对家兔离体膀胱括约肌的收缩作用,差异有统计学意义(P<0.05)。维拉帕米可以加强前列通窍胶囊含药血清对膀胱括约肌的舒张作用,但差异无统计学意义(P>0.05)。结论:随着给药天数的增加,前列通窍胶囊含药血清增加膀胱逼尿肌收缩幅度、收缩频率及活动力,降低膀胱括约肌张力的作用逐渐加强。中、高浓度含药血清组对膀胱逼尿肌、括约肌的作用差异有统计学意义。前列通窍胶囊含药血清对膀胱逼尿肌可能是通过L型钙通道来发挥作用;对膀胱括约肌可能是通过α受体来发挥作用。结果表明:前列通窍胶囊在增加膀胱逼尿肌活动力的同时,降低膀胱括约肌张力,有利于尿液排出,改善老年男性因前列腺增生症(BenignProstatic Hyperplasia,BPH)引起的逼尿肌收缩力受损后出现尿线变细、排尿等待、尿后滴沥不尽等排尿不畅临床症状。
【Abstract】 Objective: To study the effects of pharmacological serum of QianlieTongqiao Capsule on the detrusors and sphincter muscles of urinary bladder in vitroof rabbits by serum pharmacological methods and investigate the possiblemechanisms.Method: Pharmacological serum of Qianlie Tongqiao Capsule was prepared andstrips in vitro were suspended in the tissue chambers, activity of them was measuredvia Powerlab biological signalling. The effects of pharmacological serum of QianlieTongqiao Capsule in various concentration and those of various blockers on themwere observed. To investigate the possible mechanisms.Result:1、Drug-containing serums prepared after3days,5days and7days ofconsecutive feeding with Qianlie Tongqiao Capsule (3D,5D and7D serum) can allenhance the contraction amplitude, frequency and activity of detrusor, and reducetension of sphincter. Compared with the control group and the blank serum group,the difference was significant for7D serum(P <0.05). Compared with3D serum,7Dserum was significant difference in effects on bladder muscle (P <0.05).2、Compared with blank serum group,7D serum in lower, medium and highconcentration (L-, M-and H-serum) can all enhance the contraction amplitude,frequency and activity of detrusor, and reduce tension of sphincter. Compared withblank serum, the M-and H-serum both show significant difference in effects onbladder muscle (P <0.05). Compared with blank serum group, Qianlie TongqiaoCapsule-containing serum group and Longbishu Capsule-containing serum group inthe same concentration can both enhance the contraction amplitude, frequency andactivity of detrusor, and reduce tension of sphincter. And the difference wassignificant (P <0.05). The difference between the two groups was not significant(P>0.05). 3、Compared with M-serum, Verapamil can block the contraction effects ofQianlie Tongqiao Capsule-containing serum on detrusor, and the difference wassignificant(P<0.05). Qianlie Tongqiao Capsule-containing serum can reduce thecontraction effects of Norepinephrine on sphincter, and the difference was significant(P<0.05);Verapamil can enhance the relaxation effects of Qianlie Tongqiao Capsule-containing serum on sphincter, and the difference was significant(P<0.05).Conclusion: With increasing days of drug-feeding, there were increasingeffects of Qianlie Tongqiao Capsule-containing serum on enhancing the contractionamplitude, frequency and activity of detrusor, and reducing tension of sphincter. Theeffects on detrusor and sphincter from M-and H-serum groups in differentconcentration were significantly different. The effect of Qianlie Tongqiao Capsule-containing serum on detrusor may be to play a role through the L-type calciumchannel, and effects on sphincter may be to play a role through theα1receptors. Itwas demonstrated that: Qianlie Tongqiao Capsule can enhance the activity of thedetrusor and reduce the tension of sphincter, in favor of excreting urine. It cansignificantly relieve the clinical symptoms of the detrusor contraction dysfunction dueto Benign Prostatic Hyperplasia (BPH), including weak urination, slow urination anddribbling urine. etc.
