轮叶党参根化学成分及其抗炎活性的研究

【摘要】 轮叶党参Codonopsis lanceolata(Sieb et Zucc)Bentham et Hooker(Campanulaceae)是桔梗科党参属多年生缠绕性草本植物,又名羊乳、四叶参等,主要分布于我国的东北、华北、华南、西南及长江中下游地区。并在辽宁、吉林等地有大量栽培。其根作为民间草药,具有补气养阴、润肺生津、消肿排脓、以及解毒疗疮之功效。此外,在韩国、朝鲜以及日本等国家,轮叶党参还是药食两用的植物。以前的研究表明,轮叶党参根含有三萜、三萜皂苷、甾醇、以及生物碱等类型化合物。现代药理研究表明,轮叶党参具有镇静镇痛、提高学习记忆功能、抗氧化和增强免疫等作用。本文主要对轮叶党参根的化学成分及其抗炎活性进行了研究。利用大孔吸附树脂柱色谱、反复的硅胶柱色谱、并结合Sephadex LH-20和反相ODS柱色谱等分离手段,从轮叶党参根乙醇提取物中分离得到5个三萜皂苷类化合物和2个苯丙素类化合物,经理化方法和核磁共振谱学分析,确定了它们的结构。其中,3个新的三萜皂苷类化合物分别命名为:codonoposideⅠ(1)、codonoposideⅡ(2)和codonoposideⅢ(3)。4个已知化合物分别鉴定为:eclalbasaponinⅩⅢ(4)、刺囊酸(5)、甲基丁香苷(6)、以及丁香苷(7)。化合物4、6,7为首次从该植物中分离得到。对从轮叶党参根乙醇提取物中分离得到的一个富含三萜皂苷流份(Fr.4)的抗炎活性进行了研究。采用角又菜胶致大鼠足肿胀炎症模型进行测定,Fr.4显示出一定的抑制足肿胀作用[44.5%inhibition af 3h after injecting carrageenan with 500 mg kg-1(p.o.),P<0.05.]。
【Abstract】 Codonopsis lanceolata (Sieb et Zucc) Bentham et Hooker (Campanulaceae) is a perennial, twining vine widely distributed in China and cultivated in Liaoning and Jilin. The roots of C. lanceolata have been used as a herbal drug for invigorating qi to replenish yin, moistening lung and encouraging production of body fluids, subsidence of swelling and evacuation of pus, removing toxic materials and treatment of sore in China, and as a tonic crude drug and a edible plant in Korean and Japan.Previous investigations have shown that the roots of C. lanceolata mainly contain triterpenoids, triterpenoid saponins, sterols, alkaloids. It has also been reported that this crude drug has exhibited many pharmacological activities, including analgesia, antioxidation, enhancing memory and immunity.In this study, chemical constituents of the roots of C. lanceolata and its anti-inflammatory activity were investigated.The H2O-soluble fraction isolated from the ethanol extracts was chromatographed on macroreticular resin column and repeated silica gel columns, combined with Sephadex LH-20 and reverse phase ODS column chromatography to give five triterpenoid saponins, together with two phenylpropanoids. Their structures were elucidated by physico-chemical properties and spectral analysis. Among them, three new triterpenoid saponins were named codonoposide I (1), codonoposide II (2) and codonoposide III (3). Four known compounds were identified as eclalbasaponin XIII (4), echinocystic acid (5), methlysyringin (6) and syringin (7). Compound 4, 6 and 7 were isolated from this plant for the first time.Furthermore, the anti-inflammatory activity of a saponin-rich fraction (Fr. 4) isolated from the root ethanol extract was evaluated using the carrageenan induced rat hind paw edema, which displayed 44.53% inhibition at 3h after injecting carrageenan with 500 mg kg-1(p.o.), P < 0.05.
