CyclinD1,p16在肥大乳房腺体组织中的表达及意义

【摘要】 目的:探讨CyclinD1以及p16蛋白在肥大乳房乳腺组织中的表达及意义。方法:选取35例肥大乳房患者作为实验组,16例正常体积乳房患者作为对照组,术前详细询问病史和查体,根据公式[1]计算乳房体积,术后行病理切片检查,将符合要求的病例纳入样本组。将术中切取的少量乳腺组织进行石蜡包埋并切片,采用免疫组化SP法,检测乳腺组织中CyclinD1及p16蛋白的表达情况并分析二者之间的是否存在相关性。结果:35例肥大乳房标本腺体组织中的CyclinD1蛋白阳性表达率51.4%(18/35),16例正常体积乳房标本腺体组织中无一例CyclinD1呈阳性表达(0/16),两组间比较差异有统计学意义(P<0.05 )。肥大乳房组腺体组织中p16蛋白阴性表达率为77.2%(27/35),正常体积乳房腺体组织中阴性率为31.3%(5/16),两组间比较差异有统计学意义(P<0.05 )。在CyclinD1阳性表达的18例样本中,p16阴性表达者17例,阳性表达者1例,CyclinD1阴性表达的17例样本中,p16阴性表达者10例,阳性表达者7例。分析显示,CyclinD1与p16的表达呈负相关(p<0.05)。结论:①CyclinD1在肥大乳房腺体中呈高表达,p16呈低表达,提示其可能参与了肥大乳房的发生和发展。②CyclinD1和p16在肥大乳房组织中的表达呈负相关,提示二者存在着一定的调控关系。
【Abstract】 Objective: To investigate the expression and significance of the Cyclin D1 and p16 in the tissue of mastoplasia.Methods: We chose 36 cases of mastoplasia patients as the experimental group and 16 cases of nomal breast as the contol group. Preoperatively, it is required to calculate the volume of every breast according to the formula[1], ask about the disease history, and do physical examination carefully. Postoperatively, pathology examination is necessary to chose the right cases as samples. With the full consent of the patients in advance, little pieces of breast tissue from the cases is cut off, paraffin imbedded and sliced. The alteration of cyclin D1 and p16 in samples were detected using highly sensitive SP immunohistochemistry method.Results: The rate of the positive expression of CyclinD1 in mammary hypertrophy group was 51.4% (18 /35), no positive expression of CyclinD1 in the normal mammary group was detected. There was obvious significant between the two groups in a statistics basis (P< 0.05). While in mammary hypertrophy group the rate of the negative expression of p16 was 77.2% (27/35). There was also obvious significant between the two groups in a statistics basis (P< 0.05). In mammary hypertrophy group, within the 18 cases of positive expression of CyclinD1, the negative expression of p16 is 17cases, while within the 17 cases of negative expression of CyclinD1, the negative expression of p16 is 10 cases. There exists negative correlation between CyclinD1 and p16 in mammary hypertrophy group.Conclusions: There were over expression of CyclinD1 and decreased expression of p16 within mammary gland in mammary hypertrophy which may be involved in the occurrence of mammary hypertrophy. Increased expression of CyclinD1 may be in certain relation with the decreased expression of p16 in the development of mammary hypertrophy.
