eIF-4G、mTOR、Bcl-2在乳腺癌中的表达及意义

【摘要】 目的:研究真核生物翻译起始因子eIF-4G、哺乳动物雷帕霉素靶蛋白mTOR、B细胞淋巴瘤/白血病Bcl-2在乳腺癌中的表达及相互关系。方法:采用S-P免疫组化染色法检测68例乳腺癌组织、45例癌旁正常组织和20例纤维腺瘤中eIF-4G、mTOR、Bcl-2的表达。结果:(1)eIF-4G在乳腺癌组、癌旁乳腺组、纤维腺瘤组中表达阳性率分别为89.7%、20.0%和35.0%,乳腺癌组表达阳性率显著高于癌旁组和纤维腺瘤组(P<0.001)。(2) mTOR在乳腺癌组、癌旁乳腺组、纤维腺瘤组中表达阳性率分别为69.1%、22.2%和20.0%,乳腺癌组表达阳性率也明显高于癌旁组和纤维腺瘤组(P<0.001)。(3)Bcl-2在乳腺癌组、癌旁乳腺组、纤维腺瘤组中表达阳性率分别为67.6%、91.1%和95.0%,乳腺癌组明显低于癌旁组和纤维腺瘤组(P<0.05)。(4)eIF-4G、mTOR、Bcl-2在癌旁乳腺组和纤维腺瘤组中的表达阳性率均无显著性差异(P>0.05)。(5)eIF-4G、mTOR在乳腺癌中的表达阳性率与年龄、临床分期、淋巴结转移均无明显相关(P>0.05),Bcl-2的表达阳性率与临床分期、淋巴结转移无关(P>0.05),但与年龄有关,患者年龄≧50岁组的Bcl-2的表达阳性率明显低于患者<50岁组(P<0.05)。(6)eIF-4G、mTOR与ER、PR、C-erbB-2无相关关系(P>0.05);Bcl-2与ER、PR呈正相关,Bcl-2与ER的一致性更好于与PR;Bcl-2与C-erbB-2无相关关系。(7)Bcl-2和eIF-4G在乳腺癌中的表达呈正相关,eIF-4G和mTOR,Bcl-2和mTOR在乳腺癌中的表达无明显相关性。结论:(1)eIF-4G、mTOR在乳腺癌中高表达,可能与乳腺癌的发生密切相关。(2)乳腺癌mTOR、Bcl-2的检测可能为乳腺癌的靶向治疗提供依据和新的思路。(3)Bcl-2与乳腺癌细胞凋亡相关,可能受ER和eIF-4G双重调控。

【Abstract】 Objective To investigate the expression of eIF-4G、mTOR and Bcl-2 and the relationship between these three proteins.Methods Immunohistochemical method was used to detect the expression of eIF-4G,mTOR and Bcl-2 in 68 cases breast cancers,45 cases surrounding cancer-tissues and 20 cases fibroadenoma tissues.Result(1)The positive expression rates of eIF-4G in breast cancer, surrounding cancer-tissues and fibroadenoma tissues were 89.7%,20% and 35% respectively, and the positive rate of eIF-4G in breast cancer was much higher than that of the other two groups(P<0.001).(2)The positive expression rates of mTOR in breast cancer, surrounding cancer-tissues and fibroadenoma tissues were 69.1%,22.2% and 20% respectively, and the positive rate of mTOR in breast cancer was much higher than that of the other two groups(P<0.001).(3)The positive expression rates of Bcl-2 in breast cancer,cancer-surrounding tissues and fibroadenoma tissues were 67.6%,91.1% and 95% respectively, and the positive rate of Bcl-2 in breast cancer was significantly lower than that of the other two groups(P<0.05).(4)The positive rates of expression of eIF-4G, mTOR, Bcl-2 in breast carcinoma and fibroadenoma group were not significantly different. (5)The expression of eIF-4G and mTOR in breast cancer were not correlated with age,the clinical stage,or lymph node metastasis(P> 0.05).There is a relationship between Bcl-2 expression and age,no relationship between Bcl-2 expression and clinical stage,or lymph node metastases.(6) The expression of eIF-4G and mTOR were not related to the expression of ER,PR and C-erbB-2(P>0.05).But the expression of Bcl-2 and ER, PR in breast cancer were positively correlated and there was no correlated between the expression of Bcl-2 and C-erbB-2 in breast cancer. (7) The expression of Bcl-2 and eIF-4G in breast cancer were positively correlated. There was no correlated between the expression of Bcl-2 and mTOR in breast cancer,and neither was between the expression of eIF-4G and mTOR.Conclusions(1)The expression of eIF-4G and mTOR may significant contribute to the development of breast cancer. (2) The Detection of mTOR and Bcl-2 may provide a basis and thinking for targeted therapy of breast cancer. (3)Bcl-2 was related to the tumor cell apoptosis which may be under the dual regulation of ER and eIF-4G.
