针刺对急性手术创伤大鼠下丘脑CRF家族肽及其受体表达的影响

Effects of Electroacupuncture on Expression of CRF Family and CRF Receptor1 in Traumatized Rats

【摘要】 目的:通过观察针刺对急性手术创伤大鼠血清激素水平和下丘脑CRF家族肽及其受体mRNA表达的影响,探讨针刺调节急性手术创伤所导致的下丘脑-垂体-肾上腺轴功能异常的神经内分泌机制。方法:雄性SD大鼠,随机分成正常组、创伤组、正常电针组、创伤电针组。创伤和创伤电针组动物在乙醚麻醉下行剖腹探查术。创伤电针组在剖腹探查术后立即给予电针处理。正常电针组和创伤电针组选用“足三里”(ST36)和“三阴交”(SP6)穴给与电针一次,针刺参数是2/15Hz疏密波,强度1-2mA,持续30min。剖腹探查术或电针结束后30min断头处死动物,取血和下丘脑(-70℃保存)。应用特异性放射免疫法检测各组动物血清ACTH、Cort、LH和T的水平,应用RT-PCR方法观察各组动物下丘脑CRF家族肽及其受体mRNA的表达。结果:1)创伤组大鼠血清ACTH水平显著下降,和正常组相比有显著差异(P<0.05);创伤电针组大鼠血清ACTH水平显著升高,和创伤组相比有显著差异(P<0.05),与正常组比较没有明显差异;正常组与正常电针组比较无显著差异。2)创伤组大鼠血清Cort水平显著上升,和正常组相比有显著差异(P<0.05);创伤电针组大鼠血清Cort水平显著降低,和创伤组相比有显著差异(P<0.05),与正常组相比没有明显差异;正常组与正常电针组比较无显著差异。3、各组动物之间血清LH和T水平无显著差异。4)创伤组大鼠下丘脑CRF mRNA的表达降低,和正常组相比有显著差异(P<0.05),创伤电针大鼠下丘脑CRF mRNA的表达升高,和创伤组相比有显著差异(P<0.05),与正常组相比没有明显差异;正常组与正常电针组比较无显著差异。51创伤组大鼠下丘脑Ucn1mRNA的表达升高,和正常组相比差异显著(P<0.05),创伤电针组大鼠下丘脑Ucn1mRNA的表达水平下降,和创伤组相比有显著差异(P<0.05),与正常组相比没有明显的差异;正常组与正常电针组比较无显著差异。61各组动物下丘脑Ucn2、Ucn3和CRF受体1mRNA的表达均无显著差异。结论:电针可调节急性创伤导致的大鼠血清激素水平的异常,电针可能通过调整急性创伤大鼠下丘脑CRF和Ucn1mRNA的表达,进而调整急性剖腹探查所致的大鼠下丘脑-垂体-肾上腺轴功能异常。电针效应具有多环节、多靶点的特点。

【Abstract】 Objective To explore the effects of electroacupunture (EA) on the abnormal hypothalamus-pituitary-adrenal axis (HPAA) in traumatized Rats, the expression of CRF peptides family and CRF receptor1 mRNA in hypothalamus was investigated.Methods Forty male Sprague-Dawley rats were randomly divided into four groups: intact group (INT, n=10); intact with EA group (INT+EA, n=10); surgical trauma group (ST, n=10); surgical trauma with EA group (ST+EA, n=10). Dorsomyotomy were performed on the rats under anesthesia. EA was administered via two stainless steel needles in two acupoints:"Zusanli" acupoint (ST36), and "Sanyinjiao" acupoint (SP6). The stimulation was generated by an EA apparatus (Hans) and lasted for 30 min. The stimulation parameters were 2mA of density and a frequency of 2/15 Hz. At the time of sacrifice, rats were decapitated and the trunk blood (1.0 ml) of four groups were collected respectively, The plasma was separated by centrifugation at 4℃and stored at-70℃until assayed. The brains were immediate removed and the hypothalamus were dissected and stored at-70℃. Concentration of ACTH, Cort, LH and T were determined by a double-antibody RIA. Total RNA was extracted using TRIzol Regent.Results The blood ACTH concentration decreased significantly in ST (P<0.05) compared with that in INT and INT+EA, and was higher in ST+ EA (P<0.05) than in ST. There were no disparities between INT and INT+EA. The blood concentration of Cort in ST group increased significantly (P<0.05) compared with that in INT and INT+EA, and was lower in ST+EA (P<0.05). While the concentration of LH and T did not change significantly between four groups. The expressions of hypothalamic CRFmRNA in the ST rats decreased significantly compared with those in the INT rats (P<0.05), and the level in the ST+EA (?) with tho(?) In the ST rats (P<0.05). However, no disparity was found between the levels in the INT and INT+EA rats. The expressions of hypothalamic UcnlmRNA in the ST rats increased significantly compared with those in the INT rats (P< 0.05), and the level in the ST+ EA decreased significantly compared with those in the ST rats (P<0.05). There were no significant changes in the expression of Ucn2mRNA, Ucn3mRNA and CRFR1mRNA among the four groups.Conclusion Based on above, EA on"Zusanli" and "Sanyinjiao" benefits the rats with surgical trauma-lead HPA disorders.
