藏茵陈类药材中活性成分的含量测定及提取纯化工艺研究

【摘要】 中国是肝炎大国,研究肝炎防治药物是我国药物研究的主要任务之一。藏茵陈是传统的高寒藏药,属藏药八珍,在治疗肝胆系统疾病方面具有独特疗效。龙胆科花锚属植物椭圆叶花锚是我国青藏高原特有的藏茵陈原植物,其化学成分主要为口山酮类、环烯醚萜类和三萜类等。现代药理学研究表明,这些成分均具有抗肝炎活性,临床治疗急慢性肝炎疗效显著。本文系统的研究了四种龙胆科藏茵陈原植物——川西獐牙菜、印度獐牙菜、椭圆叶花锚(即藏花锚)和湿生扁蕾中有效成分的含量,并采用现代提取分离纯化方法从藏花锚中得到抗肝炎活性成分。主要工作如下:1.通过对色谱条件和系统适应性进行考察,建立了四种藏茵陈原植物中獐牙菜苦苷、当药醇苷、1-羟基-3,4,5-三甲氧基口山酮和齐墩果酸的高效液相色谱含量测定方法,测定结果显示川西獐牙菜中獐牙菜苦苷和当药醇苷的含量分别为3.12%、1.66%,藏花锚中1-羟基-3,4,5-三甲氧基口山酮的含量为0.50%,均居四种植物中对应成分含量之首。2.采用正交试验设计方法,以獐牙菜苦苷、当药醇苷、1-羟基-3,4,5-三甲氧基口山酮和齐墩果酸的转移率为评价指标,并参照各试验的出膏率,对藏花锚药材中环烯醚萜、口山酮及三萜类等活性成分进行提取工艺条件优化,并确定了最佳提取工艺参数为90%乙醇回流提取3次,每次1.5h,溶剂用量为12、10、10倍药材量。3.通过调pH水沉法使藏花锚提取液初步分离,分别得到水溶性成分(环烯醚萜类和口山酮苷类)和脂溶性成分(口山酮苷元类和三萜类)两类物质。然后,采用大孔吸附树脂法对水溶性成分进行分离纯化,分别得到环烯醚萜类和口山酮苷类成分。综上所述,本文通过对藏茵陈四种代表性原植物中的獐牙菜苦苷等主要活性成分进行含量测定,并对藏花锚的主要活性成分进行了提取纯化研究,从而为藏茵陈资源的合理开发和利用提供了科学的实验依据。

【Abstract】 Zang-Yin-Chen (ZYC) is a traditional Tibetan medicine for the treatment of hepatitis and cholecystitis. The Plant Halenia.elliptica (family Gentianaceae) is used as plant materials of ZYC. It is suggested by studying chemical constituents that iridoids xanthones and triterpenes are existed mainly in Halenia.elliptica; and it is prooved by modern pharmacological experiments that extracts of Halenia.elliptica have various biological activities.The dissertation describes the contents of active fractions in deviser gentianaceae ZYC medical materials and the isolation and purification of active parts from Halenia.elliptica by modern methods. The main research results are as follows:1.The cotents of the representative constituents of the active fractions of iridoidsxanthones and triterpenes, respectively swertiamarin swertianolin 1-hydroxy-3,4,5-methoxy xanthone and oleanolic acid, were determined through HPLC method. The quantitative methods of determination of swertiamarinswertianolin and oleanolic acid were established. The contents of swertiamarin and swertiamarin in Swertia mussotii (3.12% and 1.66%) are the highest and the content of 1-hydroxy-3,4,5-methoxy xanthone in Halenia.elliptica (0.5%) is the highest.2.With the average transfer rate of the four representative constituents and extract rate as indexes evaluated, the best extraction technology of Halenia.elliptica was optimized by orthogonal experimental design. The result is countercurrent extraction with 90% alcohol for 3 times, and each time for 1.5 hours.3.The extract was preliminarily purified through sedimenting with water and adjusting value of pH, obtaining water-solubility constituents (iridoids and xanthones) and liposolubility constituents(xanthone aglucones and triterpenes). The iridoids and xanthones were isolated and purified by macroporous adsorptive resins.In this thesis, the systematical extraction and purification of Halenia.elliptica were studied for the first time.
