寿胎丸对小鼠母胎界面SOCS1,SOCS2,SOCS3 mRNA表达的影响

【摘要】 目的:观察寿胎丸能否纠正CBA/J×DBA/2免疫流产模型小鼠子宫蜕膜Th1/Th2失衡,进而对寿胎丸保胎的作用机理进行初步阐述。方法:CBA/J×BALB/c小鼠作为正常妊娠模型组,CBA/J×DBA/2小鼠自然流产模型按妊娠顺序随机分为模型空白组、寿胎丸高剂量组和寿胎丸低剂量组,一共4组,每组各10例,通过RT-PCR实验,检测各组中绒毛组织和蜕膜组织SOCS1,SOCS2和SOCS3基因的mRNA表达变化情况。比较正常妊娠模型组和自然流产空白组之间,以及模型空白组、寿胎丸高剂量组和寿胎丸低剂量组之间SOCS基因mRNA的表达变化情况。结果:自然流产模型较正常妊娠模型,SOCS1、SOCS2和SOCS3在组织中的mRNA表达水平都下降,而且SOCS1和SOCS3在统计学意义上变化显著;在自然流产模型的模型空白组、寿胎丸高剂量组和寿胎丸低剂量组中,SOCS3的mRNA表达在高剂量组中明显高于在空白组中,且差异有显著性统计学意义,而SOCS1和SOCS2的mRNA在自然流产模型空白组和寿胎丸低剂量组中较高表达,而在寿胎丸高剂量组中低表达。结论:寿胎丸能抑制SOCS1和SOCS2基因的表达,促进SOCS3基因的表达,从而促进向Th2的分化,起到保胎的作用。

【Abstract】 Objective:Study the effect on "Shou tai pill" correcting the mice decidua Thl/Th2 disequilibrium in the CBA/JxDBA/2 immune abortion model. In addition, discuss the preliminary anti-abortional effect mechanism of "Shoutai pill".Methods:Samples were divided into four groups in the mice model experiments as following, CBA/JxBALB/c mice as the normal control group, CBA/JxDBA/2 mice randomly dividing into another three groups, such as blank model control group, high dose group and low dose group according to the pregnancy sequence, and each groups for 10 samples. Identify the RNA-level expression of the mice chorion and decidua tissues SOCS1, SOCS2 and SOCS3 genes in the former four groups through RT-PCR experiment, compare the SOCS genes RNA-level expression difference between the normal pregnancy control group and the recurrent spontaneous abortion group, and also the difference among the high doze, low doze and blank model control groups.Result:Comparing with normal abortion-prone model, the RNA level of SOCS1, SOCS2 and SOCS3 all decrease in tissues during spontaneous abortions control model, and the SOCS1 and SOCS2 are biostatistics significant; There are three groups, blank model control group, Shoutai pill high dose group and Shoutai pill low dose group, the expression of SOCS3 mRNA in high dose group are more than the blank group, and the difference is significant,. However, the difference between the low dose group and blank group are not obvious. The expression of SOCS1 and SOCS2 in the high doze group is lower, while high in the blank and low dose group.Conclusion:Shoutai pill can repress the expression of the SOCS1 and SOCS2 genes and have diverse effect to the SOCS3 genes, as a result, promote the differentiation to Th2 ending with the effect of Anti-Abortion.
