藏药PA抗肝炎有效部位的基础研究

【摘要】 PA为蔷薇科植物Potentilla anserina L.的根。在民间被长期用作营养保健品与药品。本论文研究目的在于对PA抗肝炎活性的物质基础及其有效部位的制备和质量标准进行系统的研究。主要研究内容及成果如下:1、抗肝炎有效部位中单体化合物的分离及结构鉴定:采用硅胶柱色谱等方法获得了六个单体化合物,通过理化常数测定和光谱数据分析等手段鉴定了其中三个的化学结构,分别为Ⅰ:24-deoxy-sericoside (2α,3β,19α-三羟基-齐墩果酸-28-O-β-D-葡萄糖苷);Ⅱ:anserinoside(2α,3β,19β-三羟基-乌苏酸-28-O-β-D-吡喃半乳糖苷);Ⅲ:2-oxo-pomolic acidβ-D-glucopyranosyl ester (2-羰基-3β,19α-二羟基-乌苏酸-28-O-B-D-葡萄糖苷)。2、单体化合物的保肝降酶活性研究:统计结果显示,单体化合物Ⅰ、Ⅱ、Ⅲ对小鼠CCl4引起的肝损伤具有显著的保肝降酶的活性。3、有效部位的制备工艺优化:以anserinoside含量以及生产可行性为指标,最终确定了以70%乙醇提取、经D101型大孔吸附树脂纯化获得有效部位的制备工艺流程。4、质量标准的研究:利用高效液相色谱法、比色法等技术,建立了PA有效部位原料药的质量控制标准和方法。经测定,有效部位中的总皂苷含量不得低于60%,anserinoside含量不得低于15%。本论文的创新之处在于:1、首次从该植物中分离得到了六个三萜皂苷类成分,其中一个为新化合物——anserinoside。2、首次研究了PA中的单体化合物保肝降酶的活性。3、首次将其作为抗肝炎药物进行系统的研究,完成了物质基础的确定、原料药的工艺研究和质量标准的制订工作。

【Abstract】 Potentilla anserina L. having been widely used for health care in folk. In this paper, the systemic researches on active compounds (JMS) of anti-hepatitis virus effect, including isolation of compounds and technics of preparation, were done. So, the content and outcome of studies were indicated as follow:1. The isolation and structural elucidation of chemical constituents.By repeated silica gel column chromatography and recrystallization, six constituents were obtained. On the basis of physicochemical properties and spectroscopic analysis, three structures of which were elucidated as I :24-deoxy-sericoside (2a,3p,19α-trihydroxyolean-12-en-28-oic acid-β-D-glucopyranosyl ester), II: anserinoside (2α,3β,19β-trihydroxyurs-12-en-28-oic acid-β-D-galactonopyranesyl ester),III: 2-oxo-pomolic acid-β-D-glucopyranosyl ester (2-oxo-3β, 19α-dihydroxyurs-12-en-28-oic acid-β-D-glucopyranosyl ester).2. Studies on activity of protecting liver and decreasing value of ALT.S tatistically, compounds I , II and III displayed remarkable activity of protecting liver and decreasing value of ALT on rat liver trauma by CCl4.3. Technical optimization.Considering the content of anserinoside and feasibility of technics, the technological process, extracting with 70% EtOH and purifying with D101 type macroporous resin, were confirmed.4. Methods and standards for the quality control of JMS were built.Methods and standards for the quality control of JMS were built by the technique of HPLC and colorimetry. According to the results, the total saponin and anserinoside content were determined as more than 60% and 15%, respectively. The researches have several innovations.1. For the first time, six triterpenoid saponin compounds including a new compound were obtained from Potentilla anserina L.2. For the first time, the activity of protecting liver and decreasing value of ALT were studied on compoundsⅠ,ⅡandⅢ.3. For the first time, Potentilla anserina L. was systemic studied as an anti-hepatitis virus drug.
