艾灸肾俞穴对绝经后骨质疏松模型大鼠血清B-ALP和TRAP5b影响的研究

【摘要】 目的:本研究主要采用随机对照的方法,通过观察三组清洁级SD大鼠的的差异,来探讨艾灸对绝经后骨质疏松大鼠的作用,并从分子、器官水平为中医“肾主骨”理论提供现代实验依据,为绝经后骨质疏松症(PMOP)的防治寻求一种简便、安全、切实有效的方法。方法:选择清洁级SD六月龄未孕产雌性大鼠40只,体重180-200g,首先将40只大鼠随机分为空白组和手术组,分别为5只和35只。手术3个月后从两组随机各选出5只大鼠行骨密度检查,以确定造模成功。在确定造模成功后,把手术组中剩下的30只大鼠再随机分为3组,每组10只,分别为造模空白组、西药组、艾灸组。对艾灸组大鼠的肾俞穴进行艾灸,距离大鼠肾俞学大约5厘米,悬灸大鼠肾俞穴,以施术者手背皮肤感觉温热为度,每次每穴艾灸15分钟,每天1次,五天为一疗程,然后休息两天,再继续下一疗程,连续治疗12周。西药组给予尼尔雌醇lmg/kg灌胃,使用前配成混悬液(浓度为0.167mg/ml),1次/周,其余时间给予蒸馏水10ml/kg灌胃。造模空白组每次给予蒸馏水10ml/kg灌胃。12周后,所有实验动物均断颈处死,眼球取血进行指标检测。最后数据用SPSS13.0统计软件建立数据库,进行统计分析,明确三组去势大鼠血中的骨特异碱性碱酸酶(B-ALP)和血浆抗酒石酸酸性磷酸酶(TRAP5b)的差异以及血清雌二醇水平的差异。结果:骨特异碱性碱酸酶比值比较显示,西药组和艾灸组与模型组有明显差异(P<0.01),而艾灸组与西药组比较,差别没有统计意义,血浆抗酒石酸酸性磷酸酶比值比较显示,西药组和艾灸组与模型组有明显差异(P<0.01),而艾灸组与西药组比较,差别没有统计意义。西药组和艾灸组血清雌二醇水平与模型组有显著差异(P<0.01),而艾灸组与西药组之间差异无显著性。结论:造模后去势大鼠的骨转换明显增强,血清雌二醇水平显著下降。艾灸能显著提高去势大鼠血清雌二醇水平,降低去势大鼠的骨转换的高代谢情况。

【Abstract】 ObjectiveBy using random controlling group, we did this experiment in the way of observing the differences among three groups of bioclean SD rats to discuss the effectiveness of moxibustion on postmenopausal osteoporotic rats. So that we can provide modern experimental foundation for the theory of Chinese medicine that kidney governs bones on the molecular and organic level, in the hope of establishing a simple, safe and effective way to prevent and treat PMOP.Method40 bioclean six-month-old virginal female SD rats, weighing between 180^200 grams, were randomly divided into 2 groups,5 in the blank group and 35 in the operation group. After taking away the epoophorons of rats in the operation group, we randomly chose 5 rats from each group and test the density of the whole rats’bones to make sure that the model-creating operation was successful. The remaining 30 rats in the operation group were divided randomly into 3groups:model、moxibustion and the Nilestriol group. Moxibustion to "Shenshu" points were applied to SD rats in the moxibustion group for 15 minutes every day. Each course lasted for 5 days and had a rest of 2 days. The applicable temperature should be based on the warm feeling of the experimenter’s hand. The Nilestriol group were given Nilestriol lmg/kg to fill the stomach. Confected Nilestriol into suspension, (The density is 0.167mg/ml).It was given to the Nilestriol group once a week. The other time gave the Nilestriol group distilled water 10ml/kg to fill the stomach. The Model group were given distilled water orally.After the experiment of 12 weeks, all rats were put to death and blood samples were extracted from their eyes. All the data were analyzed by the software SPSS13.0 to make clear the differences of B-ALP, TRAP5b and E2 among the ovariectomized rats of the three groups.ResultSD rats in the Nilestriol group and moxibustion group had a significant difference with Model group in B-ALP (P<0.01) and TRAP5b (P<0.01). No obvious differences have been observed between the Moxibustion group and the Nilestriol group in B-ALP and TRAP5b. E2 level in the moxibustion and model group was significantly different(P<0.01), while the contrast was not obvious between the moxibustion group and the Nilestriol group.ConclusionThe dissolution of bony tissue is notably increased in ovariectomized rats, while E2 level is remarkably decreased. This experiment showed that moxibustion can significantly increase the E2 level of ovariectomized rats and decrease the high metabolism of the dissolution of bony tissue in ovariectomized rats.
