FASCIN-1和MMP-1在喉鳞癌中的表达及其临床意义

【摘要】 目的:探讨Fascin-1和MMP-1在喉鳞状细胞癌和癌旁组织中的表达及其临床预后中的意义。方法:免疫组化SP法检测50例喉鳞状细胞癌及其癌旁0.5cm、1.0cm和正常组织中Fascin-1和MMP-1的表达情况,并结合临床随访,分析其临床病例参数及患者预后的关系。结果:Fascin-1在喉鳞癌中的表达为在癌组织、癌旁0.5cm、1.0cm及正常组织中的表达率依次降低,分别为80.0%、27.5%、22.5%、4.0%,两两比较结果显示,除了癌旁0.5组与癌旁1.0组,癌旁1.0组与癌旁2.0组的fascin-1阳性表达率差别无统计学意义之外(P值大于0.008),其余各组间差别均有统计学意义(P值小于0.008)。MMP-1在喉癌中的表达为在癌组织、癌旁0.5cm、1.0cm及2.0cm的表达率依次降低,分别为85.0%、35.0%、17.5%、5.0%,两两比较结果显示,除了癌旁0.5组与癌旁1.0组,癌旁1.0组与正常组的MMP-1阳性表达率差别无统计学意义之外(P值大于0.008),其余各组间差别均有统计学意义(P值小于0.008)。此结果与Fascin-1的表达情况类似。喉鳞癌中Fascin-1蛋白的表达与癌的临床分期,病理分化程度及淋巴结转移相关,与性别和年龄无关。MMP-1蛋白的表达同样与癌的临床分期,病理分化程度及淋巴结转移相关,与肿瘤的位置无关。喉鳞癌中二者的表达呈正相关(P<0.05)。结论:Fascin-1和MMP-1在喉鳞癌的侵袭和转移中发挥重要作用,二者的联合检测可能成为预测喉鳞癌预后的生物学指标;并为以Fascin-1和MMP-1为靶点的癌症治疗提供了思路。
【Abstract】 Objective:To study the expression of Fascin-1 and MMP-1 (matrix metalloproteinase-1) in laryngeal squamous cell carcinoma (LSCC) and adjacent tissues and their clinical significance.Methods:Using citric-SP-immunochemical technique,the expression of Fascin-1 and MMP-1 were studied in different regions of larynx squamous cell carcinoma including 40 cases laryngeal carcinoma and adjacent tissues which was 0.5、1.0 and normal tissue away from cancer.Results:The expression of Fascin-1 reduced in sequence of cancer tissue,at 0.5cm、1.0cm and normal tissue adjacent to carcinoma,which were 80.0%、27.5%、22.5%、4.0%, the results of comparison with two group indicate that,there was no significant difference in rate of addition (P> 0.008)with group of 1.0cm and 2.0 cm and the group of 0.5cm and 1.0cm.the other groups differences were statistically significant (P<0.008). the expression of MMP-1 degraded in sequence of cancer tissue,at 0.5cm、1.0cm and normal tissue adjacent to carcinoma,which were 85.0%、35.0%、17.5%、5.0%, results of comparison with two group indicate that,there was no significant difference in rate of addition (P> 0.008)with group of 1.0cm and normal tissue and the group of 0.5cm and 1.0cm.the other groups differences were statistically significant (P<0.008).the result same to expression of fascin-1.the expression of Fascin-1 protein in Laryngeal squamous cell carcinoma have a relationship with clinical stage of cancer, tumor location, tumor differentiation and lymph node metastasis,but not associated with age and gender. the expression of MMP-1 have a relationship with clinical stage of cancer, tumor location, tumor differentiation and lymph node metastasis, but not associated with age and gender.the expression of Fascin-1 and MMP-1 were positive correlation(P<0.001).Conclusion:Fascin-1 and MMP-1 maybe directly associated with the development of LSCC, which may play an important role in the process of invasion and metastasis of LSCC,United detection of Fascin-1 and MMP-1 may be an important biologica factor affecting the prognosis judgement of LSCC.
