齐聚谷氨酸多肽的合成

【摘要】 作为生物体的重要组成部分,多肽在生物、医学、化学和材料科学等领域有着重要的地位,因而有效地人工合成多肽具有非常重要的意义和价值。本论文主要描述两种齐聚谷氨酸多肽—γ-苄酯L-谷氨酸四肽和γ-异丙酰胺L-谷氨酸四肽的有效合成及其表征。采用液相多肽合成法,以L-谷氨酸为起始原料,经氨基或羧基选择性保护或脱保护和酰胺化偶联反应,有效地合成了这两种齐聚多肽,合成量达到几克级。通过优化实验条件,实现了较高的收率和光学纯度。通过1H NMR对合成中的重要中间体和目标产物分别进行了表征。

【Abstract】 As a typical bioactive matter, peptides have become interesting candidates for applications in the fields of biology, medicine, chemistry and material science. So it’s very significant and valuable to synthesize peptides effectively.In present thesis, synthesis and characterization of two oligopeptides based on L-glutamic acid were described.With highly optimized and fully reproducible solution-phase methods, y-benzyl ester-terminated L-glutamic acid tetramer and y-isopropyl amide-terminated L-glutamic acid tetramer were prepared effectively on the multigram scale by successive reactions such as selective protections of amino or carboxyl and amide coupling. Their synthetical conditions were optimized to achieve easy purifications, high yields and good optical purities.The two oligopeptides as well as some key intermediates were characterized by 1H NMR, respectively.
