两种菝葜属植物(Smilax)化学成分及抗肿瘤活性研究

【摘要】 菝葜属植物我国产60多种,目前从该属植物中已经分离出来的单体多集中在甾体皂苷,且部分具有较好的生物活性。本课题综合运用正相硅胶柱、Sephadex LH-20、大孔树脂,C18反相等色谱及波谱分析技术和化学分析方法,对两种菝葜属植物(菝葜,Smilax china L.和牛尾菜S. riparia.DC)中化学成分进行了分离纯化与结构鉴定,共分离化合物6个,其中鉴定了3个(菝葜2个,牛尾菜1个),分别为:芹菜素-7-O-a-L-鼠李糖苷(1),2-甲基-5-(1,1-二甲氧基-1-甲酰基)呋喃(2),牛尾菜酚(3),其中化合物2,3为首次报道的新化合物,化合物3为是一个骨架全新的新化合物,化合物1为首次从该属植物中得到。同时采用现代医学、生物学技术和手段,研究了提取物以及所分离的部分化合物对小鼠黑色素瘤细胞B16和乳腺癌细胞MCF-7的体外抗肿瘤活性。结果表明提取物A,G对小鼠黑色素瘤细胞B16抑制效果明显(IC50分别为2μg/mL,30μg/mL),应是菝葜和牛尾菜抗肿瘤作用的活性成分之一。所有的牛尾菜提取物在高浓度(1,000μg/mL)对对乳腺癌细胞MCF-7的增殖均起到有效抑制。推断出牛尾菜具有一定的抗乳腺癌活性。目前还有所分离的部分化合物的结构鉴定工作以及所分离化合物和部分提取物的肿瘤抑制复筛实验和肿瘤细胞凋亡实验仍在进行中。

【Abstract】 There are more than 60 species of Smilax in China. Many steroidal saponins separated from the genus and some of them show certain biological activities till now. Herein, The constituents of the rhizomes of Smilax. China L. and Smilax riparia DC. were isolated and purified by repeated silica gel chromatography, SephadexLH-20 and macroporous resin. Six compounds were obtained and four of them were identified as apigenin-7-O-rhacoside(c), 2-methyl-5-(1,1-dimethoxyl-l-formyl) furan(2), ripariaol(3) on the basis of MS, 1D and 2D NMR spectroscopic analyses and chemical evidence. Compounds 1,2 were separated from S. china. L and the other one was separated from S. riparia DC.. Compounds 2,3 are reported for the first time,expecially Compounds 3 is a compeletely novle structure,and Compounds 1 were isolated from the genus Sm ilax for the first time.Some pharmacological technologies were used to study the antitumor activity of extracts towards melanoma cells B16 and breast cancer cells MCF-7 in vitro. Extract A, G inhibited the growth of B16 cell best, their IC50 are 2μg/mL,30μg/mL respectively. The results suggested that extract A, G play important roles in the antitumor activities of S. china L. and S. riparia DC. However, all extracts of S. riparia DC. inhibited the growth of the breast cancer cells MCF-7 effectively especially at their highest concentration (1,000μg-mL-1). The result indicated that S. riparia DC. have certain bioactivities against breast cancer.The structural elucidation of some complex compounds and the anti-tumor re-screening of some phenolic constituents and extracts from S. china L. are carrying out on the way now.
