1800MHz电磁波出生前暴露对大鼠海马GFAP、NCAM及GABA受体表达的影响

【摘要】 目的了解1800MHz电磁波出生前暴露对大鼠海马NCAM和GABA受体表达的影响方法7周龄SD雌性大鼠32只,雄性16只,适应环境一周后以雌:雄=2:1交配,受孕后雌鼠随机分为4组(两个实验组和两个对照组)。实验组采用1800MHz射频电磁波对大鼠于孕0~20天进行21天连续暴露,每天12小时,功率密度为0.5 mW/cm2和1.0 mW/cm2,各实验组分别设置虚拟暴露作为对照组。暴露期间的温度、湿度、背景噪声等环境条件稳定。暴露完成后在F1代子鼠长至三周龄和七周龄,每组随机抽取8只子鼠采用免疫组织化学方法分别对海马GFAP、NCAM及GABA受体进行染色,然后采用图像分析系统分析海马CA1、CA3和DG区(齿状回)的蛋白表达。结果1.0mW/cm2暴露组未发现对三周龄、七周龄F1代大鼠海马各区GFAP、NCAM、GABA受体表达有影响,其表达与对照组无统计学意义;0.5mW/cm2暴露组三周龄F1代大鼠海马各区NCAM、GABA受体表达和七周龄GFAP、NCAM、GABA受体表达均出现了下调,其表达与对照组相比有统计学意义。结论本实验条件下,1800MHz功率密度0.5mW/cm2电磁辐射出生前暴露使三周龄大鼠NCAM与GABA受体和七周龄大鼠GFAP、NCAM与GABA受体在海马各区表达下调。

【Abstract】 Objective:To identify effects of expression on GFAP、NCAM and GABA receptor in Rat Hippocampus after prenatal exposure to 1800MHz Radiofrequency Field.methods:32 SD(Sprague Dawley) Female rats at age of 7 week-old were ran domly divided into four groups(8 rats in each) after one week for environment al adaptation and successful mating.Rats in the exposure groups were respective ly and continuously radiated between 0 days of pregnancy and 20 days of pre gnancy(12 hours/day) by 1800MHz microwave fields with power density of 0. 5 mW/cm2 or 1.0 mW/cm2.Simultaneously,two sham-controls were carried out r espectively. When the offspring were 3 week and 7week old we randomly sele cted eight pups from each group and dissected the brains.The brains tissue sect ions were performed by immunohistochemistry to demonstrate the expression of GFAP、NCAM and GABA receptor. Immunoreactivity in the hippocampus was analyzed by computer assisted image analysis system.Results:in the 0.5 mW/cm2 exposure group GFAP、NCAM、GABA receptor expression:decrease in the area of CA1、CA3、and DG of 3 week old and 7 week old rats’s hippocampus.Compared to the control groups,there was significant difference. In the 1.0 mW/cm2 exposure group GFAP、NCAM、GABA receptor expression: increase in the area of CA1、CA3、and DG of 3 week old and 7 week old rats’s hippocampus,Compared to the control groups,there was significant difference.Conclusion:there was a decrease in the expression of GFAP、NCAM and GABA receptor in the area of CA1、CA3、and DG of hippocampus in 3 week old and 7 week old rats after prenatal exposure to 1800MHz radiofrequency field with power density of 0.5mW/cm2.
