1型糖尿病患者PAF-AH活性检测及其基因单核苷酸多态性的研究

【摘要】 目的观察1型糖尿病(T1DM)病人血浆血小板活化因子乙酰水解酶(PAF-AH)的活性以及PAF-AH基因G275A多态性与1型糖尿病(T1DM)的相关性。方法选择T1DM病人115例,对照组106例,测定血浆PAF-AH活性,采用PCR—RLFP法测定PAF-AH基因G275A基因型。结果T1DM患者的血浆PAF-AH活性为(18.95±5.43)nmol.min-1.ml-1;健康对照者血浆PAF-AH活性为(26.65±3.71)nmol.min-1.ml-1,两组比较差异具有统计学意义(P<0.05)。与对照组相比,TIDM组G275A多态性位点GA基因型频率显著高于对照组(36.52%vs 22.64%,P<0.05)。结论T1DM患者的血浆PAF-AH活性降低。PAF-AH基因G275A多态性可能与1型糖尿病相关。
【Abstract】 Objective To investigate the plasma PAF-AH level in type 1 diabetes patients and the association between the PAF-AH gene polymorphism and type 1 diabetes mellitus.Methods 115 type 1 diabetes patients and 106 controls were selected to determine plasma PAF-AH by PAF-AH Assay kit,and the PAF-AH gene G275A genotypes were detected by PCR-RLFP.Results The activity of plasma PAF-AH in the patients with type 1 diabetes was significantly lower than that in the controls(18.95±5.43 nmol.min-1.ml-1 vs 26.65±3.71 nmol.min-1.ml-1,P<0.05).Compared with control group,the frequencies of the GA genotype on G275A polymorphism were significantly higher in T1DM group(36.52% vs 22.64%,P<0.05).Conclusion The activity of plasma PAF-AH in the patients with type 1 diabetes is significantly lower than controls.The G275A polymorphism of PAF-AH gene maybe related to type 1 diabebes mellitus.
