木豆叶对绝经后骨质疏松症的防治作用

The Effects of Preventing and Cure of Cajanus Cajan(L.) Millsp on Rats Ovariectomized Induced to Postmenopausal Osteoporosis

【摘要】 木豆叶具有补肾,活血化瘀的功效,能促进新骨形成和促进血管新生,这一机理正是治疗骨科疾病的关键;中医学认为绝经后骨质疏松症以肾虚血瘀为病机关键——依据“以药测证,证效相应”的理念,我们认为木豆叶可用于防治绝经后骨质疏松。研究主要考查木豆叶对去卵巢大鼠PMOP的防治作用,选取3月龄SD雌性大鼠,按体重随机分为正常组、模型组、龙牡壮骨组、木豆叶高剂组、木豆叶中剂组、木豆叶低剂组,除正常组外其余各组去势法诱导绝经后骨质疏松症模型,3天后开始防治性给药。于给药八周后大鼠眼球采血,放免法检测血浆ET水平(广州中医药大学核医学实验室),硝酸还原酶法检测血清中NO含量,ELISA法检测各组大鼠血清VEGF含量;第十周双能X法检测除木豆叶中剂量以外的所有组大鼠全身骨密度(广州中医药大学第三附属临床医院骨伤科医院),取每组5只大鼠左侧股骨制作普通病理切片。结果如下:1、全身骨密度:模型组比正常组大鼠骨密度明显减少,说明大鼠PMOP造模成功;龙牡壮骨颗粒组,木豆叶高剂组,木豆叶低剂组高于模型组,说明龙牡壮骨颗粒和木豆叶对大鼠PMOP都有防治作用。2、骨病理:正常组大鼠股骨病理切片显微镜下观察到骨小梁饱满、排列有序,模型组骨小梁多处断裂、小梁间间隙增大、甚至融合成片,龙牡壮骨颗粒组骨小梁饱满但排列不规则,部分区域小梁间间隙增大,木豆叶高、中剂组骨小梁粗壮、排列有序,木豆叶低剂组骨质均匀但骨小梁稀疏。3、ET、N0和VEGF:PMOP大鼠ET升高,NO下降,ET/NO平衡被破坏,血瘀证形成,龙牡壮骨颗粒和木豆叶均能上调ET水平,增加NO含量,抑制血瘀证形成;模型组PMOP大鼠与正常组大鼠血清VEGF并无差异,而各给药组血清VEGF均明显高于模型组,由此推测龙牡壮骨颗粒和木豆叶能促进去势后大鼠VEGF表达,促进血管生长。本实验证明PMOP大鼠全身骨密度降低,骨小梁稀疏,ET/NO失衡,与中医认为妇女PMOP以肾虚血瘀为病机关键相符;经龙牡壮骨颗粒、木豆叶防治的PMOP大鼠全身骨密度,骨小梁形态明显好于模型组,说明二者都能防治大鼠PMOP,木豆叶高剂组尤为突出;其机理可能与调节ET,NO,VEGF水平相关。

【Abstract】 Cajanus Cajan(L.) Millsp,with effects of invigorating the kidney, activating blood circulation and dissipating blood stasis,can promote new bone formation and promotion of angiogenesis the mechanism of which is the key to the treatment of bone disease.The Traditional Chinese Medicine deemde that postmenopausal osteoporosis is due to kidney deficiency and blood stasis predominantly.So basis to the idea of“definite the kind of syndrome with effective drugs,while syndrome and effects match”,we belive that Cajanus Cajan(L.) Millsp can be used for prevention and treatment of the postmenopausal osteoporosis.This study examined the effects of Cajanus Cajan(L.) Millsp on bone mineral density and bone tissue morphology of ovariectomized rats.Select the 3-month-old female SD rats which were randomly divided into normal group,model group,Longmu Zhuanggu group,Cajanus Cajan(L.) Millsp high-dose group,Cajanus Cajan(L.) Millsp middle-dose group,Cajanus Cajan(L.) Millsp low-dose group.In addition to the normal group,other groups Establish postmenopausal osteoporosis mode through ovary resection of female rats. After 3 days,begin to delivery drugs in order to prevent and treat PMOP.The cycle eye mines blood on the eighth week of administration,RIA detection for ET levels in plasma(nuclear medicine laboratory),nitrate reductase assay for NO content in serum to explore for Cajanus Cajan(L.) Millsp the role of regulation vascular endothelial function.On the tenth week,in addition to Cajanus Cajan(L.) Millsp middle-dose group,all rats of groups else were detected whole body bone mineral density by dual-energy X(in Orthopedic hospital);ELISA assay to measure the serum VEGF levels,the left side femoral of 5 rats taken in each group to produce general pathology.The results are as follows:1、whole body bone density(BMD):The model group obvious reduced,compared to the normal group,which showed that making the model of rats PMOP through ovary resection is successful;Longmu Zhuanggu group,Cajanus Cajan(L.) Millsp high-dose group,Cajanus Cajan(L.) Millsp low-dose group are higher than the model group significantly,which can explained that Longmu Zhuanggu and Cajanus Cajan(L.) Millsp for rats with PMOP has the prevention and treatment function.2、bone pathology:Under the microscope observed bone trabeculae are full and the arrangement order of normal group rats,while model group bone trabeculae fracture,increased space between trabeculae,and even integration into cavity.Longmu Zhuanggu group bone trabeculae appear to be full but with an irregular arrangement,part of the region between the gap increased. Cajanus Cajan(L.) Millsp high and middle dose groups,low-dose group trabecular bone sparse.3、ET、NO and VEGF:Rats with PMOP ET elevates and NO drops,so the balance of ET/NO is destroyed.It leads to the formation of blood stasis.Longmu Zhuanggu group and Cajanus Cajan(L.) Millsp can promote the ET level,increase the NO content,inhibit the formation of blood stasis;The model group PMOP rats and normal group rats blood serum VEGF do not have the difference,but obviously delivering drugs groups are higher than model group respectively.From those,we infer that the mechanism of Longmu Zhuanggu group and Cajanus Cajan(L.) Millsp preventing and cure rats with PMOP is to regulate the balance of ET/NO,promote VEGF to express,advance the blood vessel to grow, increase the blood vessel permeability,ensure to supply enough nourishment for bone microcirculation and growth.This experiment proved that rats with PMOP show whole body bone density reduced,bone trabeculae sparse,ET/NO unbalanced,all of that correspond with the symdrom of kidney deficiency blood stasis of woman with PMOP.Longmu Zhuanggu group and Cajanus Cajan(L.) Millsp both can prevent and cure rats with PMOP effectively.It concerns to adjust the ET,NO,VEGF level possibly.
