虎杖中白藜芦醇提取工艺研究

【摘要】 本文对中药材虎杖分别采用乙醇回流法、纤维素酶辅助提取法、微波辅助提取法和超声辅助提取法的提取工艺进行了实验研究,用紫外分光光度法检测白藜芦醇含量,计算出白藜芦醇的提取率,考察每种提取方法的工艺参数对提取结果的影响。文中对影响不同提取方法的各主要因素进行单因素实验。根据单因素实验结果,采用正交试验设计,考察了提取温度、提取时间、料液比、乙醇浓度对回流法提取结果的影响,得出提取时间对提取结果影响最为显著;考察了酶添加量、酶解温度、酶解时间、pH值对纤维素酶辅助提取结果的影响,得出酶解温度为主要影响因素;考察了微波作用功率、微波作用时间、料液比、乙醇浓度对微波辅助提取法提取结果的影响,结果表明乙醇浓度为主要影响因素;考察了超声温度、超声时间、料液比、乙醇浓度对超声辅助提取法提取结果的影响,结果表明乙醇浓度为主要影响因素。得到了四种不同提取方法各自适宜的提取工艺。在此基础上,通过综合比较四种提取方法的提取效果,得到纤维素酶辅助提取法、微波辅助提取法、超声辅助提取法均比传统乙醇回流提取法提取效率高,能耗低,可以认为是更佳的提取方法。酶法提取的最优提取条件为:酶解温度为50℃、pH值为5.0,纤维素酶与虎杖粗粉配比为1:500,酶解90min,结果比传统乙醇提取法提取率提高2倍;微波辅助醇提取白藜芦醇的最佳提取条件为:以体积分数为80%的乙醇溶液为提取剂,料液比为1:20,在510W功率下微波60s,结果比传统乙醇提取法提取率提高近3倍;超声法提取的最佳提取条件为:溶剂为乙醇,体积分数为80%,超声温度为50℃,超声时间为10min,料液比为1:20。与传统醇提工艺相比,提取率提高近3倍。

【Abstract】 Extracting techniques is experimentally studied by using methods of alcohol-extraction, cellulose-assisted extraction, microwave-assisted extraction and ultrasonic-assisted extraction for Polygonum cuspidatum. According the content of resveratrol that is extracted from Polygonum cuspidatum measured by the ultraviolet radiation method, the extracting yield is gained. And the extracting effect infected by the technical parameters of each method is discussed.Through series experiments designed according univariate experiments,According to the univariate experiments,the orthogonal experiments are carried out to analysis individually the main factors influencing the resveratrol yield, the effects of temperature, extracting time, alcohol dosage and alcohol concentration of extracting solution on resveratrol yield are studied in alcohol-extraction method. The results show that the extracting time of extracting solution is the most effective factor. In cellulase-assisted extraction experiment,the effects of cellulose amount, temperature , extracting time, and pH of extracting solution on resveratrol yield are studied .The results show that the temperature of extracting solution is the most effective factor.In microwave-assisted extraction experiment, the effects of microwave power, actuation time, alcohol dosage, alcohol concentration of extracting solution on the resveratrol yield are analyzed, alcohol concentration are found to be remarkable influence factors. In ultrasonic-assisted extraction experiment, the effects of temperature, extracting time, alcohol dosage, alcohol concentration of extracting solution on the resveratrol yield are analyzed, alcohol concentration are found to be remarkable influence factors. Furthermore the results of these experiments indicate that both grinding and pre-soakage can improve the Astragaloside IV yield considerably.By comparing extraction yield of the above four extraction methods, the cellulase-assisted extraction, microwave-assisted extraction and ultrasonic-assisted extraction method is considered as the better three than the alcohol-extraction.Gentle extraction conditions ,which were determined experimently as follows : cellulase-assisted extraction : cellulose amount 5.000g, temperature 50℃, extracting time 90min, and pH 5.0 ; microwave-assisted extraction: microwave power 510W, actuation time 60s, alcohol dosage 1:20, alcohol concentration 80% ; ultrasonic-assisted extraction: extraction temperature 50℃, extraction time 10 min and the alcohol concent ration 1:20. Compared with traditional extracting technology by ethenol ,extraction yield of resveratrol was increased by about 3 times by using this ultrasonic extraction technology.
