益康药酒的抗衰老药效学初步研究

【摘要】 制备益康药酒,以小鼠为实验对象,采用多项指标,观察益康药酒的抗衰老作用,并初步探讨其作用机理。具体实验方法为将60只小鼠,雌雄各半,随机分为六组,即益康药酒高剂量组、益康药酒中剂量组、益康药酒低剂量组、模型组、阳性对照组、空白对照组。以D-半乳糖(D-gal)所致亚急性衰老小鼠为模型,除空白对照组外其他各组均每天颈背部皮下注射5%的D—gal生理盐水溶液0.025ml/g/d,空白对照组每天在颈背部皮下注射等量的生理盐水,连续注射42天。同时灌胃给予益康药酒高、中、低三个剂量组益康药酒,剂量分别为8.8g/kg/d、4.4g/kg/d、2.2g/kg/d,阳性对照组给予维生素E(VitE),剂量为20mg/kg/d。空白对照组与模型组用等体积的蒸馏水灌胃。六周后,以跳台法及避暗法的学习记忆成绩、脾脏指数、肝组织中单胺氧化酶(MAO)以及血清中超氧化物歧化酶(SOD)、丙二醛(MDA)活性为指标,观察益康药酒延缓衰老的作用。结果益康药酒制剂能增强D—gal所致衰老模型小鼠的学习及记忆能力,增加免疫器官的重量,不同程度地提高血清中SOD的活性,降低血清中MDA和肝脏中MAO的含量。说明益康药酒具有明显的延缓衰老作用,其作用机理可能与其提高抗氧化酶活性、清除自由基和增强机体免疫功能有关。
【Abstract】 Preparation YiKang medicinal liquor, choose mice for experiment object, to observe anti-aging effect of Yikang medicinal liquor with many indexes, and mechanism of YiKang medicinal liquor on aged mice induced by D-galactose. Specific empirical method: Choose 60 six months mice and randomly divide into six groups: YK high dose group、YK medium dose group、YK low dose group、model control group, vitamine E group and blank control group. Except blank control group, the aged mice were induced by 5% D- galactose at 0.025ml/g/d for 42 days, the blank control group were induced by the same volume normal saline. Meanwhile they were treated with three doses of YK : 8.8g/kg/d、4.4g/kg/d、2.2g/kg/d, vitamine E group were treated with Vit E: 20mg/kg/d. Blank control group and model control group were treated with the same distilled water .after six weeks, test learning and memory results of jumping stand and to avoid dark experiment, detect the activity of MAO in liver and the activity of SOD、MDA in blood serum, to observe anti-aging effect of YK.YK could significantly increase the ability of learning and memory, the immune organ weight, improve the activity of SOD in blood serum, decrease the activity of MDA in blood serum and the activity of MAO in liver. YK has good anti-aging effects, which is probably due to effects of improving antioxidation, removing free radicals and enhance organism immune function.
