苦瓜种子发育过程中质量变化的基因型差异及降糖多肽-K提取、纯化条件研究

【摘要】 通过对不同基因型苦瓜品种种子发育过程中果实、种子的外部形态及生理指标的变化和降糖多肽-K提取、纯化条件的研究,从而确定种子的最佳采收时间。研究结果如下:1、对33个苦瓜种质资源的植株、果实和种子等28类形态学性状进行聚类分析,将供试资源划分为3个组群:即野生型组群(组群Ⅰ)、密瘤小果型组群(组群Ⅱ)及长大果型组群(组群Ⅲ);各类群在形态学存在明显的差异,都能体现本组群的典型形态学特征,从而反映出他们基因型存在明显的差异。各组群间存在明显的地域性差异,地域范围内的种质遗传距离较近,而地域间的种质遗传距离较远。2、对野生型、长大果型、密瘤小果型3个组群的12个苦瓜品种种子发育过程中的果实及种子的形态学性状变化及种子生活力、干重及含水量的变化进行了调查,将种子发育过程分为3个时期:快速膨大阶段、干物质积累阶段及成熟脱水阶段;各基因型的苦瓜种子在发育过程中发生规律性的变化,野生型苦瓜MC4种子发育时间比较短,在授粉后18天(18DAA)即发育到充实成熟;长大果型的MC6苦瓜种子发育时间比较长,在授粉后22天(22DAA)即发育充实成熟;密瘤小果型的MC11苦瓜种子发育时间最长,在22DAA即发育充实成熟,发育过程中果实、种子的外部形态以及种子干物质含量、含水量都发生规律性的变化,存在明显的基因型差异。3、通过酸、碱溶液浸提沉淀,薄层层析初步鉴定到降糖多肽-K的迁移率为0.288,和Khanna院士报道的0.29极为相近,可以初步确定图4-3中B点蛋白可能为降糖多肽-K。4、优化降糖多肽-K变性聚丙烯酰胺凝胶(SDS-PAGE)的条件,分离胶浓度为12%、交联度3%的变性聚丙烯酰胺凝胶能够将目标蛋白条带很好的分开。

【Abstract】 This experiment carried on changes of morphological traits, physiologic indices and the accumulate trend of polypeptide-K during seed development of 33 M.charantia.L.Ascertain best time during seed development of seed.The results as follows:1、Cluster analysis based on morphological 28 traits was carried out on 33 M.charntia germplasm accessions.Results indicated that these 33 accessions could be grouped into three major clusters,namely wild type (clusterⅠ),small fruit type(clusterⅡ)and large fruit type(clusterⅢ);The three major clusters have conspicuous differences in morphological traits,all of them reflect morphological traits.All the morphological traits reflect conspicuous differences of genetype.The genetic distances among the accessions obtained from the same ecological zone are small,while wide genetic distances could be observed among the germplasm from different ecological zones.2、Twelve germplasm accessions of M.charantia were used in this experiment to study the relationship between the changes of fruit morphological traits and the stage of seed maturity.Germination percentage, seed dry weight and seed moisture content of different seed developing stage were determined.Results indicated that seed development of M.charatia could be classified into three stages:the rapid enlarging,the dry matter accumulation and the dehydration.Regularity changes developed during seed development of genotypic M.charantia.Seed development time of wild type M.charantia shorter,seed reached filling maturity after18DAA.Seed development time of long fruit type M.charantia longer than wild type M. charantia,it take 22DAA to reach filling maturity;Seed development time of small fruit type M.charantia longest in three major clusters,seed reached filling maturity after22DAA.The changes of fruit and seed morphological traits,Germination percentage,seed dry weight and seed moisture content of different during seed development,all of them display the differrence of genotypic.3、K-polypeptide was extracted from Momordica charantia seeds with vitriol and alkali fusion and initia appreciation with thin layerchromatography (TLC).The transport ratio is 0.288 extraordinary 0.29 called Khanna.B proteinum of Figure4-3 maybe K-polypeptide.4、The condicine of Bitter melon seed polypeptide sds-page had been improved,Results indicated that 12%saprate gel and 4%stack gel could saprate Bitter melon seed polypeptide best.
