光合细菌六味地黄汤发酵工艺的优化及其主要成分的研究

【摘要】 光合细菌六味地黄汤是以优选的有益光合细菌作为菌种,加入到中药提取液中,再按照现代发酵工艺制成产品,是一种含有中药活性成分、菌体及其代谢产物的全组分发酵液的新型中药发酵加工制剂。本研究将现代生物技术与传统中药方剂有机地结合起来,将光合细菌引入到六味地黄汤中进行发酵代谢,通过光合细菌的生物转化功能和自身营养价值的利用,以强强联合的方式,挖掘出传统中药方剂的潜在药用价值。本研究首先完成了光合细菌的培养和菌种鉴定,并以光合细菌菌数为考察指标,根据其生长条件的要求,分别以六味地黄汤浓度、代谢pH值、代谢时间、接种量为因素,设计了L9(34)正交试验。由试验结果得出,No.2为光合细菌六味地黄汤制备工艺的最佳条件,为全面深入地开展光合细菌六味地黄汤的药理学和化学成分研究奠定了可靠的基础。然后从药理学方面研究了六味地黄汤经过光合细菌发酵代谢处理后的制剂对免疫功能低下小鼠的免疫增强作用,并与传统六味地黄煎剂进行疗效的比较。通过对比实验结果,验证了No.2为光合细菌六味地黄汤制备工艺的最佳条件,且与同浓度的传统六味地黄煎剂比较,更能显著地增加免疫功能低下小鼠的脾脏重量。进一步采用高效液相色谱方法,通过一系列的比较分析,可以发现经过光合细菌代谢后的六味地黄汤与代谢前的六味地黄汤相比,大部分主要成分发生了变化。且通过光谱匹配,发现其峰面积较大的色谱峰主要是来源于茱萸光合液和丹皮光合液。而且可以看出光合细菌代谢后六味地黄汤的色谱峰数比代谢前的六味地黄汤少,可能是因为经过光合细菌的代谢作用,转化成了小分子物质。因此需要通过高效液相-质谱联用技术来分析鉴定溶液中的主要化合物,这为后续工作指明了研究的方向。中药复方采用微生物发酵法进行制备,具有一般方法所无法比拟的优势,可以为开发新药、提高药物疗效、降低药物毒副作用的研究提供新的手段,为中药的发展开辟新的研究空间。进行中药发酵研究也具有成熟的现实条件,应当成为我国中药现代化研究的内容之一,从而更好地为人类服务。
【Abstract】 LW metabolized by PSB is produced by modern fermentation technics, which takes choice useful PSB as bacteria and brings them into extract of Chinese traditional medicine. It’s a new preparation of Chinese traditional medicine processed by microbial fermentation, which contains active components of Chinese traditional medicine, photosynthetic bacteria and their metabolic products.We band modern biotechnology and formula of traditional Chinese medicine together organically, and put PSB into LW initially to make a fermentation and metabolism. We want to discover its potential officinal value by means of strong-strong combination, the function of biotransformation of PSB and the utilization of themselves’ nutritive value.First of all, we finished the culture of PSB and the identification of the species. Taking the amount of PSB as the parameter, we make an orthogonal experimental design with three levels and four factors, which are the concentration of LW, pH of the solution, the culturing time and the concentration of PSB, on the basis of the environmental conditions of the growth. We can draw a conclusion that No.2 is the best of all preparation technics of LW metabolized by PSB from the results of orthogonal experimental. It lays a reliable foundation for the full and deep study of pharmacology and chemistry on LW metabolized by PSB.And then, we make a pharmacological research on the effect of LW fermented and metabolized by PSB on the immunity of rats, which are under lower immune function. We compare the traditional LW with the new preparation, which is metabolized by PSB. The findings indicate that No.2 is the best of all preparation technics of LW metabolized by PSB. It can improve the spleens’weight of rats under lower immune function, more than LW of the same concentration.Moreover, we proceed to a series of comparison and analysis by HPLC, the most principal components of LW metabolized by PSB had been changed, compared with LW before metabolized. Furthermore, we can find that the principal components which peak area is big came from Fructus corni and Cortex moutan metabolized by PSB according to spectral matching. We can also find that the peaks of LW metabolized by PSB are less than LW before metabolized. The reason may be that the components of LW had been transformed into many substances with small molecular weight after metabolized by PSB. So we need to analyze and identify the principal compounds in all of solutions by HPLC/MS, which shows the direction of the future study for us.The formula of Chinese traditional medicine, which is prepared by the fermentation of microorganisms, is so excellent that other formulas prepared by general methods can not compare with it. It can provide a new method for the study of drug development, the improvement of curative effect, reducing poisonous and side effect. It can also exploit a new investigative field for the development of Chinese traditional medicine. The study on the fermentation of Chinese traditional medicine, which has been provided with mature and realistic conditions, should be one of modern researches of Chinese traditional medicine in our country, thus it can make a better service for human beings.
