茵陈蒿黄酮提取及其β-环糊精包结物制备
Extraction of Artemisia Capillaries Thunb Flavones and Preparation of Its β-cyclodextrin Inclusion Compound

【摘要】 用五种不同方法提取茵陈蒿黄酮类化合物,其中用超临界CO2萃取法时,首次使用了碱性夹带剂。以总黄酮提取率为考察指标,优化了提取条件。研究结果表明,用超临界CO2法萃取茵陈蒿黄酮类化合物时,最佳提取条件下提取率为3.88%;“乙醇回流—固定床”法、微波辅助水法、微波辅助乙醇法、微波辅助β-环糊精水溶液法最佳提取条件下的提取率分别为3.08%,2.90%,3.09%,3.68%。五种提取方法对比可知,超临界CO2流体萃取法是一种很好的提取茵陈蒿黄酮类化合物技术,β-环糊精能显著提高茵陈蒿黄酮类化合物提取率。大孔树脂对茵陈蒿黄酮类化合物的静态吸附实验表明,中极性大孔树脂(DM-301)对茵陈蒿黄酮类化合物有较好的吸附效果,且体积分数为70%的乙醇水溶液解吸率较高。用共沉淀法制备了茵陈蒿黄酮-β-环糊精包结物,并进行了稳定性、抗氧化性和抑菌实验。研究结果表明,固定β-环糊精用量1.000 g、溶剂用量20mL,粗黄酮粉用量1.500 g、温度25℃和包结时间2h时,包结效果最好,包结物收率达75.5%;25℃时,包结物溶解度为3.20g/100 mL;以芦丁计,包结物的包结比为1:2(摩尔比,茵陈蒿黄酮:β-环糊精):包结物对超声波和氧气的稳定性较茵陈蒿黄酮类化合物有明显提高;茵陈蒿粗黄酮粉和包结物均有明显的抗氧化能力,且茵陈蒿粗黄酮粉抗氧化能力强于包结物;抑菌实验结果表明,茵陈蒿粗黄酮粉及其β-环糊精包结物对大肠杆菌、金黄色葡萄球菌、藤黄八叠球菌等三种常见细菌均有明显的抑菌能力,且包结物的抑菌能力比黄酮粉强。
【Abstract】 Flavone was extracted from Artemisia Capillaries Thunb by five kinds methods, and alkaline cosolvent was used firstly in supercritical CO2 extraction. The extracting conditions were optimized using the extracting rate of total flavonoids as indicator. Results show that the extracting rate by supercritical CO2 extraction, sorbitic-extracting-reflux-ethanol method, microwave-assisted-water method, microwave-assisted-ethanol method and microwave-assisted-β-cyclodextrin-solution method under the optimum conditions is 3.88%, 3.08%, 2.90%, 3.09% and 3.68%, respectively. Compared with four others, supercritical CO2 extraction is a very good technology for extracting flavone from Artemisia Capillaries Thunb. Meanwhile, flavone-extracting rate from Artemisia Capillaries Thunb is remarkably increased byβ-cyclodextrin.Static absorption experiment of macroreticular resin for flavonoids in Artemisia Capillaries Thunb shows that middle polarities macroreticular resin (DM-301) has high absorption ability to flavonoid compounds and 70% alcohol-water solution (v/v) delivers a high desorption effect for the flavonoids in the Artemisia Capillaries Thunb.Inclusion compound ofβ-cyclodextrin and flavone in Artemisia Capillaries Thunb was prepared by coprecipitation method, and its stability, non-oxidizability and bacteriostasis were experimentally tested. Results show that the inclusion effect is the best and inclusion yield reaches 75.5% withβ-cyclodextrin of 1.000g, solvent dosage of 20ml, virgin flavone powder of 1.500g at 25℃for 2h. The solubility of inclusion is 0.032g/mL at 25℃. According to rutin standard, when molar ratio of flavone toβ-cyclodextrin is 1:2 in inclusion compound, the stability of inclusion is greatly increased for ultrasonic and oxygen in comparison with flavone in Artemisia Capillaries Thunb. The virgin flavone powder in Artemisia Capillaries Thunb and the inclusion have evidently non-oxidizability and the former is better than the latter. Bacteriostasis tests indicate that the virgin flavone powder in Artemisia Capillaries Thunb and inclusion have evidently bacteriostasis for Escherichia coli, golden staphylococci and sarcina lutea and the latter is better than the former.
