胶质瘤中PTEN、FAK的表达及相关性的研究

【摘要】 目的:探讨PTEN、FAK在不同级别胶质瘤中的表达及其相互作用。方法:收集四川大学华西医院神经外科于2000年~2006年经开颅手术所得的50例胶质瘤临床标本;其中男23例,女27例。年龄6-73岁,平均年龄42.6岁。10例对照组正常脑组织标本,均取自我科同期行脑挫裂伤失活组织清除、内减压术的病人,其中男7例,女3例。年龄8-71岁,平均年龄41.2岁。应用SP免疫组化法,检测50例脑胶质瘤标本中PTEN、FAK的表达情况。结果:不同级别的胶质瘤中PTEN、FAK表达水平不同,PTEN在胶质瘤中总的阳性表达率是50%,其中Ⅱ级为84.6%,Ⅲ级为41.2%,Ⅳ级为35.0%。统计分析表明PTEN在Ⅱ级与Ⅲ级和Ⅳ级肿瘤之间的表达有显著的差异,而Ⅲ级、Ⅳ级之间没有差异。FAK在胶质瘤中的表达总的阳性表达率是74.0%,其中Ⅱ级为38.5%,Ⅲ级为82.4%,Ⅳ级为90.0%,统计分析表明FAK在Ⅱ级与Ⅲ级和Ⅳ级肿瘤之间的表达有显著的差异,而Ⅲ级、Ⅳ级之间没有差异。并且PTEN与FAK在胶质瘤中的表达呈负相关。结论:PTEN的阳性表达率随着胶质瘤级别的增加而降低,FAK则随着胶质瘤级别的增加而增加。在胶质瘤中PTEN表达缺失或是突变可以引起FAK的过度表达,从而增强胶质瘤的增殖和浸润。

【Abstract】 Objective: To explore the expression of PTEN、FAK in different grading gliomas and their interaction in tumor proliferation and invision.Methods: Fifty surgical specimens of gliomas from 23 males and 27 femals in Huaxi Hospital of Sichuan University during 2000~2005 were choosen. Age ranged from 6-73 years, average 42.6 years .All specimens were got from dissecting the gliomas in craniotomy. Ten controlled specimens came from the evacuation of devitalized tissue in brain contusion and laceration and internal decompression. The age ranged from 8-71 years, average 41.2 years.The sp immunohistochemical method was used to measure the expression of 50 cases glioma specimens in different grades.Results: Expression of PTEN、FAK in gliomas were varied among different grading gliomas. The general positive rate of PTEN in human gliomas was 50.0%, II grade was 84.6%, III grade wae 41.2%, IV grade 35.0%.The differences of expression level among II grade, III and IV grade were statistically significant; There was no difference between III and IVgrade gliomas. The general positive rate of FAK in human gliomas was74.0%, II grade was 38.5%,III grade wae 41.2%, IV grade 35.0%,.The differences of expression level among II grade, III and IVgrade were statistically significant; There was no difference between III and IVgrade gliomas. Additonally, PTEN expression was negatively correlated with the expression of FAK in gliomas. Conclusions: The positive rate of PTEN decreased with the development of glioma grade level, FAK increased in high grade glioma. The expression depletion or mutation of PTEN will lead to the overexpression of FAK, and promote the proliferation and invision of gliomas.
