“劳倦过度、房事不节”肾阳虚模型小鼠HPTA及舌象变化的研究

【摘要】 目的:观察“劳倦过度,房事不节”诱发的肾阳虚模型小鼠下丘脑-垂体-甲状腺轴的改变和舌象的变化。方法:选健康雄性昆明种小白鼠150只,随机分为对照组、劳倦过度组、房事不节组、劳倦过度+房事不节组(简称模型组)和金匮肾气丸组共5组,各30只。用强迫小鼠游泳法造成劳倦过度,以Colldege效应诱发雄性小鼠房事不节,用强迫小鼠游泳法造成劳倦过度并以Colldege效应诱发雄性小鼠房事不节建立肾阳虚小鼠模型,用金匮肾气丸作为治疗药物,观察各组小鼠的体征,并于第2周、4周和6周时,测定各组小鼠的体重、体温和血清TSH、T3、T4含量:石蜡切片,HE染色,光镜下观察各组小鼠的舌组织形态学改变。结果:实验第4周,模型组出现典型的肾阳虚证的表现。模型组小鼠体重、体温和血清TSH、T3、T4含量下降;舌象呈淡白舌,齿痕明显,舌背黏膜丝状乳头密度、核分裂相频数下降,上皮层厚度、角质层厚度增加。金匮肾气丸组干预治疗后,各指标有一定的改善。结论:“劳倦过度,房事不节”诱发的肾阳虚模型小鼠的一般状况与肾阳虚患者的临床表现一致,对下丘脑—垂体—甲状腺轴的影响符合临床研究和其他造模对下丘脑—垂体—甲状腺轴的影响;肾阳虚模型小鼠舌的组织病理改变以上皮增殖减慢、代谢降低为主要特征。表明采用“劳倦过度、房事不节”法可较成功地复制肾阳虚小鼠模型,为中医肾阳虚证本质提供较为理想的动物模型。

【Abstract】 Objective: To observe the changes of HPTA and the tongue signs in mice with kidney-yang deficiency model induced by "over fatigue and sexual strain" Methods: 150 healthy male mice of Kunming species were chose,and randomly divided into five groups: contrast group, fatigue group,sexual strain group,over fatigue with sexual strain group (briefly named as model group) and group treated with JinGui Shenqi Wan. There were 30 mice in each group. Kidney-yang deficiency models were induced by forcing mice to swim to make them over fatigue and inducing male mice to sexual strain with Colldege effect. JinGui Shenqi Wan was used to treat them,then the signs were observed in the second, fourth and sixth weeks,such as the weight, temperature,TSH, T3 ,T4, tongue signs in serum and tongue histological changes. Results: The mice in model group present the typical manifestations of kidney-yang deficiency at the end of the fourth weeks. Compared with the contrast group, the weight, temperature,and TSH,T3 and T4 level in serum model group obviously decreased. The tongues of the mice in model group are whitish with obvious tooth prints. Their filiform papilla density and number cell division in tongues obviously decreased. The thickness of epidermis and the thickness of stratum cornea of the tongues obviously increased. Those in JinGui Shenqi Wan group had the tendency of increasing compared with those in model group. Conclusion: The conditions of the model mice with kidney-yang deficiency due to over fatigue and sexual strain accorded with the clinical manifestations of patients with kidney-yang deficiency,the changes of HPTA accorded with clinical researches and the influence of HPTA of other models. The pathological changed of the tongue tissues in the model mice with kidney-yang deficiency were mainly characterized by lowered increasing of epidermis,decreased metabolism.
