消化道肿瘤相关基因在哈萨克族食管癌中表达改变的研究

【摘要】 目的:研究与消化道肿瘤相关的原癌基因与抑癌基因在哈萨克族食管癌组织中的表达并探讨其在哈萨克族食管癌发生、发展中的作用。方法:采用逆转录多聚酶链反应技术检测这些基因在20例食管癌组织及其相应癌旁组织中的表达情况。结果:在检测的原癌基因中20例食管癌组织与其癌旁组织相比Survivin基因表达改变明显增高的为10例,占50%;cdc42基因表达明显增高的为7例,占35%;抑癌基因表达改变不明显。与肿瘤转移相关的MMP-7基因表达明显增高的为8例,占40%。结论:Survivin和Cdc42基因表达的改变提示它们所参与的细胞凋亡及细胞周期的改变在哈萨克族食管癌的发生、发展过程中起了一定的作用。MMP-7基因表达的改变提示部分哈萨克族食管癌有转移倾向。部分标本中检测基因表达无明显改变的结果提示哈萨克族食管癌可能有其独特的发生发展机理。

【Abstract】 Objective: To study the expression alteration of intestinal related genes in carcinogenesis of Hazak’s esophageal cancer. Methods: The expression of these genes was detected in 20 cases of esophageal cancer tissues and tumor-adjacent normal tissues by the method of Reverse Transcription Polymerase Chain Reaction. Results: In detected oncogenes, the expression of Survivin gene increased in 10 cases compared with that in tumor-adjacent normal tissues within 20 cases; 7 cases show higher expression level for cdc42; The expression of tumor suppress genes does not show dramatic changes in all detected samples. There are 8 cases have increased expression level for MMP-7 that related with tumor metastasis. Conclusion: Changes of Survivin and Cdc42 gene expression indicate that they might play certain roles in carcinogenesis and progression of Hazak’s esophageal cancer. The results of MMP-7 gene expression indicated that part of Hazak’s esophageal cancer have metastasis inclination. The results that the expression level of all detected genes does not show dramatic change in a group of samples suggest that the specific mechanism may control the development for Hazak’s esophageal cancer.
