As2O3对NB4细胞PTTG和c-myc基因表达的影响

【摘要】 目的观察三氧化二砷(As2O3)对NB4细胞垂体瘤转移基因(pituitary tumor transforming gene,PTTG)和C-myc基因表达的影响,为As2O3治疗APL提供新的试验依据。方法自苏州大学医学院购买人急性早幼粒细胞白血病NB4细胞株,精心培养,所有试验用细胞均为对数生长期细胞。分别用0.5、1.0、2.0μmo1/L的As2O3作用NB4细胞24h,48h,根据As2O3干预的时间和浓度不同分组,同时设立不加As2O3干预的NB4细胞组并选取SMMC-7721肝癌细胞系作为阳性对照组,每组均设三个复孔,收集各组别细胞用TRIzol法提取总RNA后立即进行RT-PCR,PCR产物在1%琼脂糖凝胶上电泳,凝胶电泳图像应用Biosens gel image analysis system进行PTTG和C-myc基因表达强度的半定量分析。结果未加As2O3的NB4细胞对照组和SMMC-7721肝癌细胞阳性对照组PTTG和C-myc基因的表达强度比较差异无显著性(P >0.05);0.5、1.0、2.0μmo1/L的As2O3作用NB4细胞24h,48h后PTTG基因的表达强度逐渐减弱,与未加As2O3的NB4细胞对照组比较差异有显著性(P <0.05)。C-myc基因的表达强度亦逐渐减弱,与未加As2O3干预的NB4对照组比较P <0.05。结论PTTG和C-myc在NB4细胞中也有高表达,提示PTTG和C-myc的高表达在急性早幼粒细胞白血病( acute promyelocytic leukemia,APL)发病过程中可能起着重要作用,As2O3的下调NB4细胞PTTG和C-myc的表达作用可能是其治疗APL的分子机制之一,其具体机制尚待进一步研究。

【Abstract】 Objective Observes the arsenic trioxide (As2O3) to affect the NB4 cell pituitary tumor transforming gene (PTTG) and the C-myc gene expression influence, is As2O3 treats APL to providethe new experimental basis. Methods From Suzhou University purchaser acute promyelocytic leukemia NB4 cell line, carefully cultivating, All test cells were used to grow a few cells. Separately uses the different density As2O3 to affect the NB4 cell for24 hours, 48 hours, According to the As2O3 intervention time and the density differentgrouping, simultaneously sets up does not add As2O3 the interventionthe NB4 cell group, Selects the SMMC-7721 liver cancer cell line to take the masculinecomparison group, each group supposes three duplicate holes, Collects each group of other cells to withdraw total RNA after theTRIzol law to carry on RT-PCR immediately, The PCR product the electricity swims on 1% agrose gelatin, Results Has not added As2O3 the NB4 cell comparison group and SMMC-7721 livercancer cell masculine gender comparison group PTTG and the C-myc geneexpression intensity comparison difference does not have Significant (P> 0.05); after 0.5、1.0、2.0μmo1/L As2O3 affects NB4 cell 24h, 48h the PTTG gene expression intensity gradually weakens, with has not added As2O3 the NB4 cell comparison group comparison difference to have Significant (P <0.05)。The C-myc gene expression intensity also gradually is weaken, with hasnot added As2O3 the intervention the NB4 comparison group to compare . P <0.05。Conclusion PTTG and C-myc also have the high expression in the NB4 cell, prompts PTTG and the C-myc high expression arises in the process in APL possibly to play the vital role, As2O3 decline NB4 cell PTTG and the C-myc expression function possiblyis it treats one of APL member mechanisms,Its concrete mechanism also needs further to study.
