成熟期烟叶表面蔗糖酯的研究

【摘要】 蔗糖酯是烟草中一种重要的香味前体物质,由腺型茸毛分泌产生。本文以云烟85、K326、K346、G80、NC89、NC82、红花大金元(本论文中简称为红大)7个烤烟品种为试验材料,采用高效薄层色谱扫描法,对各烤烟品种叶表面蔗糖酯进行了定性和定量分析,并研究了各烤烟品种成熟期不同叶位烟叶表面蔗糖酯与其他化学成分的相关性和成熟进程中烟叶表面蔗糖酯及其他化学成分的动态变化。同时,采用自然雨淋和盖膜处理的方法,分析比较了雨水对烟叶表面蔗糖酯的影响。主要研究结果如下:(1)建立了分离测定烟叶表面蔗糖酯的薄层色谱扫描分析法的试验条件,优化了流动相溶剂和染色剂的配比。通过对不同流动相的层析效果进行比较,确定按甲苯—乙酸乙酯—甲醇—水(体积比为10:5:4.5:0.2)为展开剂,脲—磷酸—正丁醇混合为显色剂分离测定烟叶蔗糖酯的效果最好。为快速分析检测烟叶表面蔗糖酯创造了试验条件。(2)对7个烤烟品种打顶后不同叶位烟叶表面蔗糖酯的分泌量及其与其他化学成分的相关性进行了分析。结果表明:不同烤烟品种表面蔗糖酯总量存在明显的差异,且各叶位烟叶表面蔗糖酯含量差异呈现相似的变化趋势。其中以NC89含量最高,其次是云烟85,最低为G80。叶位之间比较发现,品种间蔗糖酯总量均呈现上部叶>中部叶>下部叶的变化规律。相关性分析结果表明,不同品种之间蔗糖酯含量差异显著。蔗糖酯与可溶性总糖、还原糖和烟碱之间呈正相关,与非还原糖和钾之间呈负相关。(3)研究了烤烟成熟过程中烟叶表面蔗糖酯及其它化学成分的动态变化。结果表明:打顶后28d内,不同烤烟品种的上部和中部叶片表面蔗糖酯含量均呈上升趋势,28~35d之间开始缓慢回落;各烤烟品种在成熟进程中蔗糖酯的变化趋势与总糖、还原糖和烟碱的变化趋势基本一致,与钾呈相反的变化趋势。(4)采用自然雨淋和盖膜处理的方法,研究自然降雨对红花大金元和K326烟叶表面蔗糖酯及其他化学成分总含量的影响。研究结果表明,雨水冲刷对烟叶表面蔗糖酯有较明显的影响。中部叶表面蔗糖酯受雨水冲刷的影响比上部叶大;K326烟叶表面蔗糖酯受雨水冲刷的影响要大于红花大金元;降雨对可溶性总糖、还原糖、非还原糖、烟碱和钾的含量影响较小。

【Abstract】 The sucrose ester is a kind of important fragrance precursor material in the tobacco,by the gland cilia secretion production. This article take the Yunyan85, K326, K346, G80,NC89, NC82,Hong HuaDaJinYuan, 7 flue-cured tobacco varieties as experiment samples,using the highly effective thin layer chromatograph scanning method to carry onqualitative and the quantitative analysis to the flue-cured tobacco leaf tobacco surfacesucrose ester; Studying different leaf position leaf tobacco surface sucrose ester and otherchemical composition relevance for the different genotype flue-cured tobacco at the sametime; Researching leaf tobacco surface sucrose ester and other chemistry target dynamicchange of flue-cured tobacco in different mature course,At the same time, using themethod of natural rain and tectorial membrane to analyze and compare rain waterinfluence upon the leaf tobacco surface sucrose ester. The main findings are as follows:(1) In order to create the condition for the fast examination and the analysis of leaftobacco surface sucrose ester, adopting thin layer chromatograph scanning analysismethod that using the highly effective thin layer chromatograph scanning methodestablishment separation appraisal leaf tobacco surface sucrose ester. After trying twodifferent flowing direction, discovering that the leaf tobacco sucrose ester separationeffect is good that taking the toluene-ethyl acetate-methyl alcohol-water (volumeproportion is 10: 5:4.5:0.2) as the developing solvent, the urea-phosphoric acid-normalbutyl alcohol mix is the developer. The thin layer chromatography’s operation is simple,the sensitivity is quickly high, accurate reliable, the reproduction quality is good, theresult is stable, and it can separate the flue-cured tobacco sucrose ester effectively, and isextremely suitable for the mass sample’s analysis, which may be a effective method forthe flue-cured tobacco sucrose ester analysis.(2) At the same time carries on the sample to 7 different genotype flue-curedtobacco after pinching off, determining different genotype roasts the leaf tobacco surfacesucrose ester secretion and analyzing it with other chemistry target relevance. The resultsindicated: Different genotype flue-cured tobacco surface sucrose ester total quantity isobviously different. The difference of various position leaf tobacco surface sucrose estercontent presents the similar change tendency, the NC89 content is highest, next is theYunyan85, the lowest is G80. Comparing the leaf position discovered upside that thevariety of sucrose ester total quantity presents the leaf>middle the leaf>lower partleafs rule. The relevant analysis result indicated that the difference of different variety ofthe sucrose ester content is remarkable, but the different leaf position differs from. The sucrose ester and the soluble total sugar, the return to original state sugar and the nicotineare positive correlation, and it is negative correlation with the non-return to original statesugar and the potassium.(3) In the flue-cured tobacco mature process, dynamic change of the leaf tobaccosurface sucrose ester and other chemistry target research indicated that middle leafsurface sucrose ester content in the 0-28d time section assumes the rise basically after tippruning,the tendency, in 28-35d starts to recede slowly after tip prtming; Different varietyin mature process sucrose ester’s main change tendency of sugar, return to original statesugar, and nicotine are basic consistent, assumes the opposite change tendency with thepotassium.(4) Using the method of the natural rain the tectorial membrane to research naturerainfall take effect on Hong Hua Da Jing Yuan with the K326 leaf tobacco surfacesucrose ester and other chemistry target total content. The findings indicated that the rainwash has a great influence on the leaf tobacco surface sucrose ester content. Comparingthe leaf position, middle of the leaf receives affects is bigger than the upside leaf.Comparing variety, K326 receives affects must be bigger than HD, and affects most about21 d after pinching off. Rains influences on the sucrose reduced when the later period ofleaf tobacco mature. The influence of rainfall to the soluble total sugar, the return tooriginal state sugar, the non-return to original state sugar, the nicotine and the potassiumcontent is small, the variety difference is not so great.
