抗生素对坛紫菜配子体生长和代谢的影响

【摘要】 以坛紫菜为试材,在不同浓度抗生素存在的条件下,通过对配子体重量、叶绿素含量和脯氨酸含量的生理测定,研究了4种抗生素对其配子体的生长和代谢的影响。结果表明,在非致死浓度下,坛紫菜配子体对氨苄青霉素和硫酸庆大霉素表现出了很强的耐受性,并且在低浓度情况下,硫酸庆大霉素对配子体的生长还表现出了促进作用。因此,这两种抗生素可作为坛紫菜配子体无菌化培养的除菌剂配合使用。而卡那霉素和壮观霉素对坛紫菜配子体表现出了很强的抑制和毒害作用,并且卡那霉素的毒性要大于壮观霉素,在培养液中加入较高浓度的卡那霉素和壮观霉素后,配子体迅速死亡,褪色变绿,重量减轻。由于坛紫菜配子体对卡那霉素和壮观霉素非常敏感,这两种抗生素可作为坛紫菜基因工程实验中有效的选择压力。本项研究在坛紫菜配子体培养的基础上,研究了抗生素对其配子体生长和代谢的影响,找出了它们的毒害浓度范围,为坛紫菜配子体培养和无菌化处理过程中避免其毒害提供了依据。同时,为坛紫菜配子体转基因研究中转化细胞的筛选找出了适宜的抗生素种类及其浓度,为坛紫菜配子体发育调控的研究开阔了思路和提供了方法。

【Abstract】 The Porphyra haitanensis is a nutritious and delicious alga. In the Europe and USA it is called Laver, and in the Japan it is named Nori. There are about 70 species of the Porphyra in the world. Now the main species of the artificial culture are P. yezoensis and P. haitanensis. The P. haitanensis is an important economic alga. So there are many researches to learn more about the alga. In the theses, the sdudy is focus on the effects of antibiotics on the growth and physiological processes of P. haitanensis.The study investigated the differences of growth and metabolism of the gametophytes of P. haitanensis treated with the four different antibiotics under different concentrations respectively by messuring the weight, the chlorophyll and proline contents. The culture condition for the gametophytes was 25℃, 2000lx, and 12:12h light:dark duration. The experimental results showed that the growth and metabolism of the gametophytes were not obviously affected by the amppicillin and gentamicin sulfate, when the concentrations of the two antibiotics were lower than 0.01mol/L. With increase of the concentration, interestingly, gentamicin sulfate promoted the growth of the gametophytesat at concentration of 0.01mol/L. The other two antibiotics, kanamycin and spectinomycin, obviously inhibited the growth and metabolism of the gametophytes when their concentrations were higher than 0.01mol/L. The inhibited effect of kanamycin was stronger then spectinomycin. So we might concluded that the amppicillin and gentamicin sulfate could be used for producting the aseptic gametophytes and the kanamycin and spectinomycin might be applied as selection pressure in the P. haitanensis genetic engineering.
