大蒜素对棘阿米巴超微结构的影响

【摘要】 目的:实验研究大蒜素对棘阿米巴超微结构的影响。方法:用无营养琼脂培养基从土壤中分离棘阿米巴,用PYG培养液培养。将棘阿米巴用10ug/ml、100ug/ml、1000ug/ml、10000ug/ml组的大蒜素处理不同浓度的大蒜素处理,对照组加等量的PYG,在28℃培养24h,在透射电镜下观察其超微结构损伤情况。结果:浓度为10ug/ml组滋养体转化为包囊,未观察到超微结构的损伤;100ug/ml组对棘阿米巴的超微结构产生轻度破坏,胞膜完整,部分线粒体出现变性、肿胀;1000ug/ml以上组棘阿米巴的结构产生严重破坏,可观察到胞膜发生皱缩破坏,胞质疏松,细胞器严重损伤,线粒体肿胀,空化,嵴断裂或消失,内外膜结构不清,部分核膜双层结构不清晰,胞质内空泡消失,部分可见调亡小体。结论:大蒜素浓度100ug/ml以上时可损伤棘阿米巴超微结构,对棘阿米巴有杀伤作用。
【Abstract】 Objective:To detect the effect of Allitridium against to Acanthamoeba spp. In vitro.Methods:A strain of Acanthamoeba was isolated from soil and cultured in PYG. The Acanthamoeba was cultured with 10ug/ml, 100ug/ml, 1000ug/ml, 10000ug/ml difference concentrations of Allitridium influence of Allitridium on the morphological feature, and ultrastructures was tested.Results: At concentration of 10ul/ml Allitridium forced the Acanthamoeba trophozoites change to. At concentrations of 100ul/ml Allitridium, we were detected light lesion that degeneration and tumefaction of mitochondria. The Allitridium concentration was more than 1000ul/ml, that Allitridium effectively destroyed the ultrastructures of the Acanthamoeba.Conclusions:At concentration of 1000ul/ml Allitridium was effectively destroying Acanthamoeba trophozoites and cystes.
