西洋参果的化学成分研究

【摘要】 本研究是建立在对珍贵资源五加科人参属植物西洋参(Panax quinquefolium L.)的果实进行广泛的资源和文献考察的基础上,采用大孔吸附树脂、层析法等现代与传统提取、纯化、分离、鉴定技术方法,从西洋参果中分离得到15个化合物,经理化常数测定和波谱数据解析确定了14个化合物结构,分别是:20(R)-原人参二醇[20(R)-protopanaxadiol,20(R)-PPD,Ⅰ]、24(R)-拟人参皂苷元[24(R)-Ocotillol,Ⅱ]、20(S)-人参二醇[20(S)-panaxadiol,20(S)-PD,Ⅲ]、20(R)-原人参三醇[20(R)-protopanaxatriol,20(R)-PPT,Ⅳ]、β-谷甾醇(β-sitosterol,Ⅴ),胡萝卜苷(daucosterine,Ⅵ),20(S)-人参皂苷-Rh2[20(S)-ginsenoside-Rh2,Ⅶ]、人参皂苷-Rg1(ginsenoside-Rg1,Ⅷ)、20(R)-人参皂苷-Rg3[20(R)-ginsenoside-Rg3,Ⅸ]、拟人参皂苷-F11(pseudogmsenoside-F11,P-F11,Ⅹ)、人参皂苷-Rd(ginsenoside-Rd,Ⅺ)、人参皂苷-Rc(ginsenoside-Rc,Ⅻ)、人参皂苷-Rb1(ginsenoside-Rb1,ⅩⅢ)、人参皂苷-Rb3(ginsenoside-Rb3,ⅩⅣ)。其中20(R)-PPD(Ⅰ)、24(R)-Ocotillol(Ⅱ)、20(S)-PD(Ⅲ)为首次在西洋参果中发现。研究了人参皂苷-Rd在大鼠体内的代谢,即大鼠分别ip 250mg/kg和po 400 mg/kg,采用薄层色谱法(TLC),高效液相法(HPLC)对收集的粪便、尿液进行检测分析,结果表明:人参皂苷-Rd在大鼠体内的代谢为人参皂苷-Rg3、F2、Ph2、coumpound K(C-K)和20(S)-原人参三醇[20(S)-PPT]。

【Abstract】 Fifteen compounds were isolated from the fruits of Panax quinquefolium L. cultivated in China on the basis of the widespread investigation of the resource and literatures with such techniques as D101 macroporous absorption resin (MAR), chromatography, and octadecylsilane (ODS) ect.. Fourteen compounds were identified according to physico-chemical properties and spectral data. Their structures were identified as: 20(R)-protopanaxadiol(I), 24(R)-Ocotillol ( II ), 20(S)-panaxadiol (Ⅲ), 20(S)-protopanaxatriol (Ⅳ),β-sitosterol (V), daucosterine (Ⅵ), 20(S)-ginsenoside-Rh2 (Ⅶ), ginsenoside-Rg1 (Ⅷ), 20(R)-ginsenoside-Rg3 (Ⅸ) , pseudoginsenoside-F11(Ⅹ), ginsenoside-Rd (Ⅺ), ginsenoside-Rc (Ⅻ), ginsenoside-Rb1 (ⅩⅢ), ginsenoside-Rb3 (ⅩⅣ). As a result, compoundsⅠ,Ⅱ,Ⅲwere isolated from the plant for the first time.The metabolism of ginsenoside-Rd in rats after ip 250 mL/kg and po 400mL/kg respectively, was studied in the paper. In the pilot study, thin-layer chromatograph (TLC) and high performance chromatography (HPLC) were used to assay the samples of the rats’ urine and feces. The results showed that ginsenoside-Rd had 5 main metabolites, which were ginsenoside-Rg3, F2, Rh2, Coumpound K (C-K) and 20(S)-propopanaxadiol.
