共轭亚油酸对前体脂肪细胞增殖和分化的影响

【摘要】 共轭亚油酸(conjugated Linoleic Acid,CLA)是一种新型功能性油脂,具有特殊的生理功能人们对CLA进行了大量的研究,发现它具有抗癌活性,降低动脉粥样硬化,调节免疫,促进动物生长,抑制机体脂肪沉积,营养重分配作用以及影响骨骼形成等多种生物学活性。目前的研究主要集中在脂肪细胞增殖与分化过程中调控生脂过程的具体分子事件和细胞事件。我国对脂肪细胞的研究刚刚起步,大部分是关于不同种属的脂肪细胞形态学的研究。本文对共轭亚油酸来源结构和生理功能、脂肪细胞来源、研究脂肪细胞分化的体外系统、脂肪细胞分化过程、分化过程中细胞形态、瘦素、脂联素的变化以及影响脂肪细胞增殖与分化的因素作一综述。本试验通过前体脂肪细胞原代培养,采用细胞生物学基本方法,观察了大鼠和艾维因鸡前体脂肪细胞分化过程中的形态变化。通过添加不同浓度的CLA到前体脂肪细胞中,应用油红O染色OD值测定,细胞染色,Leptin、adiponectin的检测等多种方法,探讨CLA对脂肪细胞增殖和分化的影响。实验结果如下:1.适合本试验室前体脂肪细胞合适接种密度为:大鼠4×10~4和鸡8×10~4.2.油红O染色表明,CLA处理组比对照组细胞数量明显减少,CLA处理组前体脂肪细胞呈梭形或不规则的多边形,细胞内脂滴较小,数目较多,细胞体积小。对照组细胞大体呈圆形,胞内脂滴较大,数量较少,胞核处于细胞边缘。3.大鼠前体脂肪细胞的倍增时间为65.5h,前体脂肪细胞接种后潜伏期约为48h左右,培养6d后前体脂肪细胞增加了3倍。前体脂肪细胞接种后的第3-9d是前体脂肪细胞的指数增殖期,第9d后前体脂肪细胞的生长进入平台期。艾维因鸡前体脂肪细胞接种后潜伏期约为48h左右,培养6d后前体脂肪细胞增加了3倍。4.实验表明不同浓度的共扼亚油酸能显著降低大鼠前体脂肪细胞的数目,而不同浓度的共扼亚油酸在降低大鼠和艾维因鸡前体脂肪细胞的数目上也有显著差异。说明CLA抑制大鼠和艾维因鸡体脂肪细胞的增殖与分化。5.而本实验中CLA处理大鼠和艾维因鸡后脂肪细胞leptin、adiponectin的分泌没有显著变化。CLA对脂肪细胞分泌功能的影响及其作用机制有待进一步研究。

【Abstract】 The conjugated linoleic acid is one kind of new function fat,had the special physiological function.CLA have the anticancer activeness,can cut down the atherosclerosis,adjusts immunity,promote animal growth,suppresses organism fatty deposition,and heavily affects the nutrition assignment,as well as affects the skeleton forming and many kinds of other biological activeness.The present research mainly concentrates in the preadipocyte multiplies and differentiation in the process to regulate the fresh fat process the concrete molecular event and the cell event.Our country just started to the fatty cell research,majority of was about the fatty cell morphology research which not homogeneous was.The experiment explored the culture condition of the preadipocyte in the given lab,on the basis of this,using biology essential method,observed the shape change in preadipocyte differentiation of rat and AA broiler.In this study,the effects of CLA on proliferation and differentiation of preadipocyte is observed by counting cell, examining leptin the adiponectin level in blood,determining OD after Safranin O staining and so on,through adding different dosage of CLA to the preadipocyte.Experimental result as follows:1.The Suitable evaccination density of preadipocyte in this test is rats:4×10~4 and chicken:8×10~4.2.The results of Safranin O staining indicated that,the cell number obviously degraded by CLA treatment.The preadipocyte in CLA treatment group assumes the irregular polygon,the fat drop smaller in the cell,and the cell volume is small.The preadipocyte in control group assumes the circular,the fat drops were bigger in the cell,but the quantity of fat drops is less than that in CLA treatment group.3.The doubling generation time of rats preadipocyte is 65.5h,and the latency period of preadipocyte after vaccination is about 48h,and the preadipocyte increase to 3 times after raised 6d.The 3-9d after preadipocyte vaccination is the doubling generation time,after 9d the growth of preadipocyte enters the balcony period.The latency period of chicken preadipocyte after vaccination is about 48h,and the preadipocyte increase to 3 times after raised 6d.4.The experiment indicated that the CLA be able to degraded significantly the preadipocyte proliferation in rats and chicken.And the different dosage of CLA had the significant deviation. 5.The secretion of leptin and adiponectin in adipocyte of rats and chicken were not striking changed after CLA treatment in this experiment.The effects of CLA on the fatty cell secretion and its possible mechanism of action need further studies.
