抗蚜苜蓿品系SSR体系的建立及遗传多样性分析

【摘要】 以九个不同抗蚜级别的苜蓿品种及四个杂交F1代为试材,摸索SSR反应体系中各个影响因素的浓度和用量,确定适合苜蓿抗蚜虫基因定位的SSR反应体系:其中Taq DNA聚合酶0.2μL(5U/μL),10×Buffer(Mg2+Plus) 2.5μL,dNTP Mixture 2μL (各2.5mM),引物各1.5μL (10μM),模板DNA1μL,ddH2O 16.3μL补足25μL。利用该体系进行扩增所得谱带清晰、稳定。利用SSR标记对不同抗蚜级别的苜蓿品种间及四个杂交F1代与其亲本间的遗传多样性进行分析,结果表明:抗蚜品种M8和HA-3具有较近的亲缘关系,二者的相似系数为0.8550,而品系S7和X2亲缘关系较远,相似系数最小,为0.5362。高抗品种HA-3与低抗品种Hunter River的相似系数为0.7391,其遗传背景差异性相对较大。此外,亲本金皇后(♂)与G3(♀)之间的遗传距离为0.5108,X2(♂)与G3(♀)之间的遗传距离为0.7156,亲本之间的遗传差异比较大。自然杂交F1代及人工杂交F1代同父母本之间的相似系数也只在0.3556~0.6333之内,与父母本有一定的遗传差异。
【Abstract】 Nine different aphid-resistant alfalfas and four cross bred F1 were used in this experiment to investigate better dosage in SSR technique system and to seek the best combination among factors.At last,the best SSR technique system was established. The Taq DNA polymerase was 0.2μL(5U/μL),the 10×Buffer(Mg2+Plus)was 2.5μL , the dNTP Mixture was 2μL ( 2.5mmol/L) , the primer was 1.5μL (10μmol/L),the template DNA was 1μL,the ddH2O was 16.3μL ,and the whole system was 25μL.The amplification bands were clear and standard by using this system. SSR markers were used to investgate the Genetic diversity of the nine different aphid-resistant alfalfa,the four cross bred F1 and their parents.The conclusions were: aphid-resistant alfalfa M8 and HA-3 were near relatives ,their similar coefficient was 0.8550.The S7 and X2 had the smallest similar coefficient that was 0.5362.The similar coefficient between high-resistant aphid alfalfa HA-3 and low-resistant aphid alfalfa Hunter River was 0.7391,they had large genetic differences of their background.Besides this,the genetic distance between Golden Kaiserin(♂)and G3(♀)was 0.5108,and the genetic distance between X2(♂)and G(3♀)was 0.7156, the genetic diversity between male parent and female parent was large comparatively.The similar coefficient between cross bred F1 and their parents was from 0.3556 to 0.6333,they had some genetic differences with their parents.
