脑络通粉对局灶性脑缺血的保护作用的实验研究

【摘要】 目的通过动物试验研究,探讨新药“脑络通粉”对大白鼠局灶性脑缺血模型损伤的保护作用,验证脑络通粉对缺血性脑中风的保护作用。方法通过血管内线栓阻断法建立大鼠大脑中动脉局灶性脑缺血模型,观察脑络通粉对脑缺血大鼠的行为状态、脑缺血面积、脑组织形态学的作用。采用八道生理记录仪观察脑络通粉在给药前后对麻醉犬血流动力学和脑血流量、脑血管阻力的作用;采用二磷酸腺苷(ADP)诱导法,用LBY-NJ型血小板聚集仪,测定60s、180s、300s血小板聚集强度,最大聚集强度(MA);采用XSN-12型体外血栓形成仪测定血栓的长度、湿重和干重;动物均皮下注射肾上腺素(Adr)0.08ml/100g,大鼠浸入冰水内5分钟造成大鼠血瘀模型,检测全血比粘度(高、中、低切变率),血浆比粘度,红细胞压积(HCT);用MC-4000型半自动血凝分析仪进行凝血酶原时间(PT)及部分凝血活酶时间(APTT)测定。另外,采用断头法记录小鼠全脑死亡的喘气时间。结果研究结果表明:脑络通粉1.6g/kg组可明显改善造模大鼠脑缺血后的神经症状(P<0.05)、缩小脑缺血范围、抑制了神经细胞的病理学改变。脑络通粉可增加麻醉犬脑血流量(P<0.05、P<0.01、P<0.001),降低脑血管阻力(P<0.05、P<0.01)。脑络通粉1.6g/kg、0.8g/kg、0.4g/kg三组剂量均对模型大鼠血浆血小板聚集具有不同程度的抑制作用(P<0.05);脑络通粉1.6g/kg组对大鼠全血比粘度的高切、中切、低切、红细胞压积与模型对照组相比,均有明显的降低作用(P<0.05、P<0.01、P<0.001);脑络通粉各剂量组对大鼠体外血栓的长度,湿重和干重、均具有明显的降低作用(P<0.05、P<0.01、P<0.001);脑络通粉1.6g/kg、0.8g/kg剂量组对大鼠部分凝血活酶(APTT)时间具有明显的延长作用(P<0.05、P<0.01、P<0.001)。另外对断头小鼠存活时间有明显延长作用(P<0.05、P<0.001)。结论脑络通粉能够改善脑循环,增加缺血区脑血流量,对脑缺血有一定的保护作用。

【Abstract】 Objective To explore the protective effect of Naoluotong Powder (NP) on the focal cerebral ischemia injury model of rat. To Verify the effect of ZP on ischemic stroke protection.Methods Focal cerebral ischemia modcl was made via the thread-occluding method to explore the protective effects of NP on the score of neurological deficits,the rate of cerebral infarction and histomorphology of cerebral ischemia in rats. The effects of ZP on the haemodynamics,cerebral blood flow and cerebral vascular resistance in the anaesthetized dog were investigated in the 8-channel physiology recorder.The ADP-induced platelet aggregation was respectively measured by LBY-NJ-platelet aggregation apparatus,The intensity of platelet aggregation and the maximal aggregation intensity in the time of 60s. 180s, 300s were Determined, The thrombus length, wet weight and dry weight were determined by the extracorporeall XSN-12 type thrombopoiesis instrument. The whole blood viscosity at high shear rate. middle shear rate and low shear rate. hematocrit and blood plasma viscosity were checked in the animal model which was made by equal hypo of Adrenalin (Adr) 0.08 ml/100g. 5 minutes bring about rat blood within rat infusion ice water stagnating. PT and APTT were Determined by MC-4000 type semiautomatic blood curdle analytical instrument. The time of mouse entire brain death was recoderd by the time recording.Results The results of research are indicate that NP was perthsed to the stomach of rats by 1.6, 0.8g/kg respectively,which could distinctly improve the score of neurological deficits of cerebral ischemia rats(P<0.05),reduce the rates of cerebral infarction rats and the level of cerebral ischemia injury,It is able to improve the cerebral tissue of pathological. It also increase the anaesthetized dog’s cerebral blood flow (P<0.05、P<0.01、P<0.001)and reduce cerebral vascular resistance(P<0.05、P<0.01 ).The 1.6 g/kg. 0.8 g/kg. 0.4 g/kg three species dosage without exception have inhibitory action to model rat blood plasma blood platelet aggregation(P<0.05). The 1.6 g/kg group has an obviously effect to reduce the whole blood viscosity at high shear rate,middle shear rate,low shear rate and hematocrit compared with model group(P<0.05、P<0.01、P<0.O01). Every dosages of NP have the obvious lessening effect to the thrombus length, wet weight; dry weight(P<0.05、P<0.01、P<0.O01).It could increase the time of the mouse takes a breath(P<O.05、P<O.O1).Conclusion NP is able to improve brain circulation, increase blood flow of cerebral ischemia aera. It has thc certain protection effect to the focal cerebral ischemia.
