肝脂消煎剂对脂肪肝大鼠肝脏CD14/LBP mRNA表达的影响

【摘要】 目的:通过对脂肪肝大鼠肝脏脂多糖受体CD14mRNA、脂多糖结合蛋白LBPmRNA表达及肝脏病理检测,观察中药肝脂消煎剂对脂肪肝大鼠的治疗效应,并探讨其作用机制。方法:采用高脂饮食和白酒灌胃,经8周建立脂肪肝大鼠模型,造模成功后用肝脂消煎剂治疗,并设东宝肝泰对照组。治疗6周后取肝组织。观测:(1)肝脏Kupffer细胞CD14mRNA;(2)肝组织LBPmRNA;(3)肝组织病理。结果:①高脂饮食加白酒灌胃8周可建立以慢性脂肪肝、脂肪性肝炎为主要病理表现的大鼠脂肪肝模型。②肝脂消煎剂能降低大鼠肝脏Kuppfer细胞CD14mRNA表达水平,其作用明显优于东宝肝泰组(P<0.01)。③肝脂消煎剂能降低大鼠肝脏LBPmRNA表达水平,其作用明显优于东宝肝泰(P<0.01)。④肝脂消组肝脏炎症计分显著降低,同时肝组织病理结果显示,肝脂消煎剂具有良好抗脂肪肝,治疗脂肪性肝炎作用,明显优于东宝肝泰(P<0.05)。结论:肝脂消煎剂治疗脂肪肝的效应及药效学机制为:1、减轻肝细胞炎症反应;2、抑制内毒素血症引起的肝脏Kupffer细胞CD14mRNA及肝脏LBPmRNA表达。
【Abstract】 Objective:To observe the therapeutic effect of Ganzhixiao apozema and approach its mechanism of action by detecting the expression of liver’s CD14 mRNA and LBP mRNA, and hepatic tissue’s pathological section in FLD Rats.Methods:High fat diet and distillate spirit were lavaged to the rats for 8 weeks to make the fatty liver rat model. Then rats were divided into four groups: normal control group, model control group, Ganzhixiao apozema treated group and Dongbaogantai treated group. After 6 weeks’treating, we obserbved the following markers: (1)CD14mRNA of Kuppfer’s cell; (2)LBPmRNA of hepatic tissue; (3) pathological section of hepatic tissueResults:①Fatty liver rat model was erected by feeding high fat diet and lavaging distillate spirit for 8 weeks, whose patho-appearance mainly was chronic fatty liver and fatty hepatitis.②Expression of CD14mRNA of Kuppfer’s cell was obviously depressed by Ganzhixiao apozema, and this effect was superior to Dongbaogantai’s(P<0.01).③Expression of LBPmRNA of liver was obviously depressed by Ganzhixiao apozema, and this effect was superior to Dongbaogantai’s(P<0.01).④Infl activity Accumulated points of liver degraded obviously in Ganzhixiao apozema treated group,and observation results of pathology of liver showed that Ganzhixiao apozema can take effect of anti-fatty liver, and treating fatty hepatitis(AH), and the effect was superior tO Dongbaogantai’s (P<0.05).Conclusion:The pharmacodynamics and therapeutic function of Ganzhixiao apozema decocting prescription to fatty liver as the following:①Lightening the inflammatory reaction of hepatic cell;②Inhibiting the expression of CD14 mRNA of Kuppfer’s cell and LBP mRNA of liver that caused by endotoxemia.
