二氢吡啶类衍生物的合成研究及大青叶等药材中三种有效成分的测定

【摘要】 本文分为两部分,第一部分是二氢吡啶类新衍生物的合成研究;第二部分是用HPLC法测定大青叶、板蓝根及青黛三味中药中有效成分色胺酮、靛蓝、靛玉红含量并建立新质量控制标准的研究。二氢吡啶是一类具有重要生物活性的化合物,其作为一种钙离子拮抗剂主要用于心脑血管等疾病的治疗;也可作为辅酶NADH模型化合物,用于仿生不对称还原。第二章研究在室温下用固相研磨法,两步合成了NADH模型化合物2,6-位未取代的1,4-二氢吡啶类化合物9个,其中5个为新化合物,并用元素分析、~1HNMR、IR等进行了结构表征。第三章以廉价的薯蓣皂素为起始原料,通过开环、氧化、水解三步反应合成中间体双烯醇酮醋酸酯。并以其为原料设计了三条路线,试图合成含甾体母核的新二氢吡啶类衍生物,进行了一些探索性研究。大青叶、板蓝根及青黛为三味常用、同源中药,一般以有效成分靛蓝、靛玉红的含量作为质量控制标准。本文利用HPLC法同时测定了大青叶、板蓝根、青黛甲醇提取和水煎处理后浸出液中色胺酮、靛蓝与靛玉红的含量,结果大青叶、青黛的甲醇提取液和水煎液都含有上述三种物质,而板蓝根只有甲醇提取液中有靛蓝和靛玉红,建立了评价这三味中药质量控制的新方法。此外,还针对实验中靛玉红出现的怪异色谱现象,通过薄层色谱、HPLC及分子极性计算等对其色谱行为进行研究。

【Abstract】 This dissertation has two parts, the first part is the study of synthesis the new dihydropyridine derivatives. The second part is the using HPLC method to determine the contents of three active components in three traditional Chinese medicine (TCM) and establish the new quality control standard.Dihydropyridine compounds are a kind of important calcium channel blockers for the treatment of cardiovascular and cerebrovascular diseases. It also used as a Coenzyme NADH model compounds for biomimetic asymmetric reduction. Nine 1, 4-dihydropyridine compounds without 2, 6-substituted were synthesized by two-step reactions. Five of which were new compounds. In the third chapter the cheap diosgenin was used as starting material to synthesize the new asymmetric 1, 4-dihydropyridines. Thus, the ring-opening, oxidation, hydrolysis reactions of diosgenin were carried out and intermediate 16-DPA was obtained. Furthermore, three novel routes were designed to synthesize the new dihydropyridine derivatives which including steroidal structure starting from 16-DPA.Folium Isatidis, Radix Isatidis and Indigo Naturalis are three commomly used traditional Chinese medicine. And the contents of their active components indigo, indirubin are generally used as quality control standard. The contents of tryptanthrin, indigo and indirubin in three traditional Chinese medicine were determined by HPLC method. The results showed that all three chemical substances are existed in Folium Isatidis and Indigo Naturalis; but only indigo and indirubin are in the Radix Isatidi’ methanol extraction. In addition, the strange chromatographic behavior which found from the experiment was studied by TLC, HPLC and molecular polarity.
