金银花指纹图谱研究

【摘要】 通过查阅大量文献资料,本文开篇对目前国内外中草药的研究概况、金银花的理化性质和药理研究以及中草药质量控制现状作出了综述性总结。本文实验部分首先根据《中华人民共和国药典》一部之规定,分别对19批金银花药材的化学成分作出了定性鉴别,依次检查了它们的水分、灰分、酸不溶灰分及其中的Cu、Pb、Cd有害元素的含量,并采用高效液相色谱法对19批金银花中绿原酸和木犀草苷的含量进行了双波长同时检测,在此期间对仪器和方法的精密度、重现性以及药材供试品溶液的稳定性作出了考察。目前,我国对中药材的质量控制仅限于对药材进行定性鉴别和常规考察,部分药材对其中的一两个成分进行了含量测定,现代医学认为,这种质控方式不能全面、完整的体现被控药材的质量,不能保证药材在临床应用中的安全性和有效性。针对于此,本文应用高效液相色谱法结合《中药色谱指纹图谱相识度评价系统2004版》软件对金银花指纹图谱进行了尝试性研究,希望能以此填补目前中药材质控方面的不足。通过建立的对照指纹图谱对金银花样品药材进行了相似性对比,由对比得出的相似度判断金银花样品质量(合格与不合格),并对影响相似度的诸多因素进行了详细讨论。

【Abstract】 The herbal medicine, as a part of Chinese traditional culture, has presented the tremendous contribution to human’s physical health. It is noted by many nations of the world for its characteristics such as abundant resource, unique impact, and less toxicity.It is the rapidly increasing development and uniting of the world economy, which trended to take new choice and challenge to Chinese herbal medicine. After taking part in WTO, although China has been providing a powerful support with deposited the abundant source, only 5 percent of export of China holding to the total world exports which is radically beyond the Japan, European and United State, and what is the reason? While the reasons came from various respects, the fundamental reason are for the long time lacking of strict quality control standard, for the coalescing with the international commerce and for the appreciated by international rules.Nowadays, the quality control in China is just checking quality or separate component, which absents comprehensive checking method merely by checking which component is existence and how much component existed in the survey and as the determinant standard to confirm whether or not eligibility of the drugs. The process puts the checking result to a dilemma between the illegal using and deserting, therefore, to solve this situation and the promotion of export of the Chinese herbal medicine,we must offer an innovation to change the flawed checking means and chart out a feasible and comprehensive quality control standard which could take more ratio of Chinese export from the world total export.Firstly, the experimentation in this thesis made some quantity discrimination to 19 batch of honeysuckle which is coming from different provinces where are planting honeysuckle. Secondly, the experimentation tests their content of water, draff, undissolved draff from acids and toxicity. Thirdly, using the HPLC mensurates the content of chlorogenic acid(CHA) and Luteolin-7-0-β-D-glucoside.Nowadays, the technology of Fingerprints Atlas has been applied as an advanced means which indeed is used in many countries and is accepted by international regulation, whereas the application of this technology in China is in the premature phase. In 2000, the State Food and Drug Administration had issued a regulation named“the Requiring of Fingerprints Atlas Technology of Injection in Chinese Traditional Medicine”which will provide a reliable evidence for the research of Fingerprints Atlas Technology in the China and illuminate the direction for the promotion of this technology. This distertation, which uses HPLC and scheme from“Evaluation Standard System to the Similarity of Fingerprints Atlas in the Chinese Traditional Medicine(2004)”, prosecutes some researches in Fingerprints Atlas of honeysuckle which include: (1) the selection of chromatogram condition which made some selection and confirmation of various samples by different distill means, distill solvent, the quantity of entered sample, temperature, flowing velocity, checking spectrum wave, the proportion of flowing phases, and method of rushing; (2) methodology which is a research method by calculating the time and RSD of relative area preserved from the peak number of chromatogram, and to assess the apparatus’accuracy and recurring and stability; (3) erecting Fingerprints Atlas which utilizes the calculating results by the program of“Evaluation Standard System to the Similarity of Fingerprints Atlas in the Chinese Traditional Medicine(2004A)”to construct the dactylogram figure by the data of HPLC of 15 batch of honeysuckles, and deduce to a complete instituted dactylogram figure; (4) mensurating of samples which is by the technique of collating the dactylogram figures and sample figures from the instituted figures which are deduced from“Evaluation Standard System to the Similarity of Fingerprints Atlas in the Chinese Traditional Medicine(2004B)”and determine the eligibility; (5) the discussing of results which includes discussing the similar impact by checking waves, fleeting time, experiment method and apparatus system; (6) suggestion and prospect which is aim at nowadays some problems in the Fingerprints Atlas and provide some innovational suggestions, furthermore prospect to the development of Fingerprints Atlas technology.It has to by incessant advancement and innovation that every technology to be obtained a full-fledged situation and ultimately to be popularized, so as the Fingerprints Atlas technology. We believe by enormously large of experiment research—herbal theory and advanced analytic technology—the Fingerprints Atlas technology will obtain a brilliant accomplishment in the domain of the Chinese herbal quality control.
