中药复方CM活性成分和中药青黛的研究

【摘要】 本论文分为两个部分,第一部分对中药复方CM生物活性部位进行了追踪和筛选,确定了中药复方CM的活性部位,并研究了活性部位的化学成分。第二部分对中药青黛进行了研究,确定了从青黛中提取靛玉红的最佳方法和溶剂,并对中药青黛的化学成分进行了分离。主要内容如下:中药复方CM水提物由桂林三金股份有限公司中药研究所提供,本论文对其生物活性部位及活性成分进行了追踪筛选和研究。将中药复方CM水提物以溶剂萃取、大孔树脂分离、硅胶柱层析分成多个部位,通过抗血小板凝聚试验筛选出了两个具有显著活性的部位:正丁醇萃取物和大孔树脂10%乙醇洗脱的部位。其中正丁醇萃取物,在浓度为0.1660g/mL时,血小板最大聚集率为3.06%,10%乙醇洗脱部位,在其浓度为0.1249g/mL时,血小板最大聚集率为6.76%。利用多种分离技术对CM活性部位的化学成分进行分离,得到3个三萜皂甙化合物,其中1个为四糖甙,1个为五糖甙,另1个为六糖甙。通过对3个化合物分别进行酸水解,确定了3个化合物的甙元结构都为常春藤甙元,同时确定了糖的种类为阿拉伯糖、鼠李糖和葡萄糖。为了确定糖链在甙元上的连接方式,对其中2个皂甙(化合物A、C)进行了碱水解,分别得到二糖甙A’和四糖甙C’以及葡萄糖,从而确定了酯糖链的结构都为两个葡萄糖。进一步应用现代波谱技术(FAB-MS,IR,1H-NMR,13C-NMR,COSY,HSQC,HMBC,TOCSY)最后确定了3个三萜皂甙的结构。它们分别为:3-氧-α-L-吡喃鼠李糖基(1→2)-α-L-吡喃阿拉伯糖基-常春藤甙元-28-氧-β-D-吡喃葡萄糖基-(1→6)-β-D-吡喃葡萄糖酯甙(化合物A); 3-氧-β-D-吡喃葡萄糖基(1→3)-α-L-吡喃鼠李糖基(1→2)-α-L-吡喃阿拉伯糖基-常春藤甙元-28-氧-β-D-吡喃葡萄糖基-(1→6)-β-D-吡喃葡萄糖酯甙(化合物B); 3-氧-β-D-吡喃葡萄糖基(1→4)-β-D-吡喃葡萄糖基(1→3)-α-L-吡喃鼠李糖基(1→2)-α-L-吡喃阿拉伯糖基-常春藤甙元-28-氧-β-D-吡喃葡萄糖基-(1→6)-β-D-吡喃葡萄糖酯甙(化合物C)。青黛是一种传统的中药材,靛玉红是其中的主要活性成分,为了从青黛中提取靛玉红,我们通过紫外分光光度法比较了青黛中靛玉红在常用溶剂中的含量,确定了其最佳提取方法及溶剂为:青黛盐酸处理后,用乙酸乙酯提取。最后对青黛中的化学成分做了初步的分离,从中分离出5个化合物,其中3个生物碱,

【Abstract】 This thesis includes two parts. In the first part, the bioactive fractions of Compound Chinese herbs of CM were studied by means of activity track. Then the chemical constituents from the bioactive fractions were isolated. In the second part, one Chinese traditional medicine named Qingdai (Indigo naturalis) were investigated. The Optimum extraction method and solvent of extracting Indirubin from Qingdai and the chemical constituents of this medicine were studied. The main contents of this thesis are as follow:The water extract of Compound Chinese herbs of CM was provided by Chinese medicine institute of Guilin SanJin Pharmaceutical Co.,Ltd. During the course of investigation, the water extract was fractioned with different polar solvents, macroporous resin and silica gel chromatography. Both n-butanol parts and 10% ethanol parts of D-101 macroporous resin were proved to have pharmacological activities against platelet aggregation through experiment on rat’s blood plasma in vitro. The results showed that n-butanol parts had exhibited obvious effect on inhibiting platelet aggregation which maximum aggregative rate was 3.06% at the concentration of 0.1249g/mL, while the maximum aggregative rate of 10% ethanol parts of D-101 macroporous resin were 6.76% at the concentration of 0.1660g/mL.To search bioactive constituents from Compound Chinese herbs of CM, we systematically investigated the chemical components of 10%~30% ethanol parts of D-101 macroporous resin. As a result, 3 compounds of triterpenoid Saponins were isolated by various means of separation, The acid hydrolysis of them yielded the same hederagenin and arabinose, rhamnose, glucose. The alkaline hydrolysis of A and C yielded the same glucose together with A’from A and C’from C, indicating their ester glycosidic linkage were two glcose. The structures of 3 triterpenoid Saponins were further identified on the basis of IR, FAB-MS, 1H-NMR, 13C-NMR and 2D-NMR (COSY, HSQC, HMBC, TOCSY) spectra, to be 3- O-α- L- rhamnopyranosyl (1→2) -α- L- arabinopyranosyl- hederagenin- 28- O-β- D- glucopyranosyl (1→6)-β- D- glucopyranosyl ester (A), 3- O-β- D- glucopyranosyl (1→3)-α- L- rhamnopyranosyl (1→2) -α- L- arabinopyranosyl- hederagenin- 28- O-β- D- glucopyranosyl (1→6)-β- D- glucopyranosyl
