金花茶叶中黄酮成分的提取与分离

【摘要】 金花茶(Camellia chrysantha( Hu) Tuyama)系山茶科、山茶属、金花茶组植物。广西特有的国家一级保护植物。具有清热解毒、利尿消肿、预防肿瘤的功效。金花茶除用于观赏外,现已应用于医药、饮料。前人对金花茶的研究主要集中在育种等领域。很少有人对其叶进行研究。本论文主要从一下四个方面对金花茶叶进行了研究。  1.对金花茶叶的茶用价值进行了研究。首先对金花茶叶中的化学成分进行了定性分析。通过定性分析,发现金花茶叶中含有氨基酸、茶多酚、黄酮类化合物等。通过茚三酮比色发,以及茶多酚类物质能与亚铁离子形成紫色络合物,利用分光光度计分别在570nm和540nm处,对它们的含量进行了测定。测得金花茶叶中氨基酸含量为 5.80%;茶多酚含量为9.66%。   2.测定了金花茶叶中黄酮含量。金花茶叶通过蒸馏水煮沸,经过大孔吸附树脂吸附,75%的乙醇洗脱,旋干,得金花茶叶子粗黄酮,在用无水酒精精制,得精制黄酮。通过外标法定量分析,测得黄酮粗品中总黄酮含量为:2.33%;黄酮精品中总黄酮含量为:4.62%。   3.利用RP-HOLC法,采用保留时间定性,发现金花茶叶中含有黄酮甙元:山奈酚和槲皮素,并通过硅胶柱层析分离出单体。经过多次硅胶柱层析和梯度洗脱,从金花茶叶中分离出了3个黄酮甙。通过各种光谱和化学方法,证实其中一个黄酮甙是山奈酚-3,7-O-α-L-二鼠李糖甙,一个可能是(-)-儿茶素-4-β-D-吡喃半乳糖甙,还有一个有待进一步研究确定。还从金花茶叶中分离出了一种三萜化合物齐墩果酸。   4.用正交实验法对用大孔树脂富集金花茶叶黄酮的工艺进行了研究。以金花茶精制总黄酮为考察指标,采用L9(34)交试验设计,考察样品液浓度、样品液流速、洗脱液乙醇浓度、树脂类型等因素对金花茶总黄酮含量的影响。确定最佳工艺条件和参数:D-101型号

【Abstract】 Jinhuacha [Camellia chrysantha(Hu) Tuyama],is a Sect.ChrysanthaChang, Theaceae,Camellia plant,It is the First - grade State protection and plant endemic species of Guangxi.It has clearing away heat and toxic material、inducing diuresis to reduce edema and prevention of tumor.Now it is used widely in manufacturing medicine, beverage and flavoring expect as a view and admire plant. Predecessors mainly are focused on breeding and Jinhuacha leaves are researched very few. This essay studies Jinhuacha leaves from the flowing four aspects.1. Studied the tea value of Jinhuahca leaves. Jinhuacha leaves contains amino acid、tea polyphenons、flavonoids and such like by qualitative analysis. Amino acid content is determined by ninhydrine colorimetry. Tea polyphenons and ferrous ion can form purple complex, its content is determined by spectrophotometer in 570nm and 540nm. The result shows amino acid content is 5.80%,tea polyphenons content is 9.96%.2. Determined the flavone content of Jinhuacha leaves.The coarse flavone is extracted from jinhuacha leaves by boiling water, collected by resin adsorption, elutriated by 75% ethanol.The refined flavone is obtained by absolute alcohol purification. The content of total flavonoids is measured by external standard method. The result shows that the content of coarse flavone is 2.33% and the content of refined flavone is 4.62%.3. Quercetin and kaempferol are found from jinhuacha leaves by their Retain-time in RP-HPLC. Their monomers are gained by silica gel column chromatography. Three flavonol glycosides and a terpenoid are isolated from Jinhuacha leaves with gradient elution, many times silica gel column chromatography and recrystallization. The structures of the terpenoid and two flavonol glycosides are verified by different kinds of spectrum and chemistry methods. They respectively are kaempferol-3,7-O-α
