补肾化瘀解毒复方对肺癌耐药逆转作用及机理研究

【摘要】 目的:通过观察中药补肾化瘀解毒复方提取物对肺癌实体瘤及肺癌耐药细胞的作用,以期能为中药补肾化瘀、解毒散结理论治疗肺癌以及逆转肺癌化疗耐药性提供客观的实验依据。 方法:以动物移植性Lewis肺癌为模型,观察中药补肾化瘀解毒复方提取物与化疗药物顺铂(DDP)联合运用,对荷瘤动物的抑瘤和增效作用。以人肺腺癌耐药细胞A549/DDP为研究对象,分别采用细胞毒实验(MTT)、流式细胞仪(FCM),观察补肾化瘀解毒复方荷瘤动物含药血清对肺腺癌敏感细胞A549和耐药细胞A549/DDP的细胞毒作用,以及对细胞周期的影响和对DDP的耐药逆转作用,补肾化瘀解毒复方含药血清对肺癌耐药细胞A549/DDP多药耐药相关蛋白(MRP)、拓扑异构酶Ⅱ(TopoⅡ)和谷胱甘肽S转移酶(GST)表达的影响,同时观察与维拉帕米(VPL)合用对肺癌耐药性的协同逆转作用。 结果:补肾化瘀解毒复方对动物移植性Lewis肺癌有显著的抑制作用,并能提高DDP对Lewis肺癌的抑制率。补肾化瘀解毒复方含药血清对A549和A549/DDP均有一定的细胞毒作用,补肾化瘀解毒复方含药血清能显著降低A549和A549/DDP细胞S期的细胞比例下降(P<0.05),进入G1期细胞数增加,进入S期细胞数下降。补肾化瘀解毒复方含药血清能显著抑制肺癌耐药细胞MRP和GST的表达,提高TOPOⅡ含量及其活性,并且补肾化瘀解毒复方含药血清与维拉帕米均能逆转A549/DDP的耐药性,而且联合使用具有更好的协同效果。 结论:补肾化瘀解毒复方对动物移植性肺癌具有抑瘤和化疗协同增效作用,能阻滞肺癌细胞进入增殖期,从而发挥抑制癌细胞生长的作用;并能增强DDP的敏感性和对DDP的耐药逆转作用,其作用机理与中药复方补肾化瘀解毒方抑制耐药细胞耐药基因MRP和GST-π的表达和提高TOPOⅡ含量及其活性密切相关。

【Abstract】 Objective:Through observing the influence of Bushenhuayujiedu(BSHYJD) recipe to solid tumor and drug resistance lung cancer cell of DDP in lung ,we can provide objective experimental evidence for the TCM theory of enhancing kidney function & transforming stasis and resolving toxic &dissipating binding in the therapy of lung cancer and reversion multidrug resistance of lung cancer chemotherapy of Bushenhuayujiedu recipy.Methods:The model of Lewis transplanted animals were applied to study the effects of tumor inhibition and synergism to tumor-bearing animals when Chinese herbs Bushenhuayujiedu recipe combined with chemotherapeutic agents Cisplatinum(DDP). We take the lung cancer resistance cell A549/DDP for research object to observe the cytotoxicity effects of BSHYJD recipe drug-serum to the lung cancer sensitivity cell A549 and lung cancer resistance cell A549/DDP, and the influnence to cell cycle and the resistance reversion to DDP. These were determined by MTT assay.
