樟子松针叶活性成分研究

Study on the Bioactive Constituents of the Needles of Pinus Sylvestris L.

【摘要】 樟子松(Pinus sylvestris L. var. Mongolica Litvin)为松科松属裸子植物,属于高大常绿乔木,分布于黑龙江和内蒙古东部大兴安岭山区及海拉尔一带沙丘地区。其针叶能祛风,活血,明目安神,解毒止痒,其主要化学成分为萜类和挥发油,黄酮及其苷类等。前期实验表明,樟子松针叶石油醚提取物和醋酸乙酯提取物具有较强的抗肿瘤作用和免疫抑制作用。为寻找具有生物活性的化学单体,本文对樟子松针叶的乙醇提取物进行了较为充分的有机化学成分研究。实验采用多种现代分离技术(包括快速层析法,凝胶色谱法和制备HPLC),对樟子松针叶各提取部分进行分离,从中共分离到50个单体化合物,通过1H-NMR,13C-NMR,2D-NMR,高分辨质谱等波谱解析方法鉴定了其中26个化合物的结构。其中3个为新化合物: 15-乙基-18-松叶酸甲酯(1);15-Acetoxy-labda-8(17),13E-dien-18- al (2) ; 18-Hydroxy-labd-8(17),13E-dien-15-acetate (3)。1个新天然产物:15-Acetoxy-labd-8(17)-en-18-oic acid (4)。已知化合物中,化合物18α-Acetoxy-labd-8(17)-en-15-oic acid (8) , 3β-Hydroxy-8(17),E-13-labdadien- 15-oic acid (10),去氢松香酸(11),7α-羟基去氢松香酸(12),7β-羟基去氢松香酸(13),7α,18-二羟基去氢松香醇(14),海松醇(16),海松酸(17),异海松醇(18),异海松酸(19),7-oxo-13-epi-pimara-8,15-dien-18-oic acid (20),α-杜松醇(21),T-杜松醇(22),eudesm-11-en-4α-ol (23),桉油烯醇(24),oplopanone (25),β-葫萝卜苷(26),均为从樟子松中首次分离得到。在前期实验证明樟子松提取物具有明确的抗肿瘤作用的基础上,以Hela细胞和A549细胞为研究对象,考察分离鉴定的单体化合物的抗肿瘤作用。研究结果表明,在实验剂量下,化合物12的细胞毒作用相对较强,对两种细胞株均有细胞毒作用。化合物7和13对Hela细胞的生长有一定抑制作用,化合物14和15对A549细胞的生长有一定抑制作用。本文研究了樟子松提取物对小鼠淋巴细胞增殖转化的抑制作用。研究结果表明,在实验剂量下,樟子松针叶石油醚提取物和醋酸乙酯提取物,表现出较强的免疫抑制活性。但各单体化合物的初步免疫抑制活性实验结果均未显示明确的淋巴细胞转化抑制作用,与原石油醚和醋酸乙酯提取物显示的较强免疫抑制作用不相对应,有必要对樟子松的免疫抑制化学成分进行深入研究。

【Abstract】 Pinus Sylvestris (Pinus L. ) is a kind of large green arbor year occouring mainly in Dongbei Province, China. The needles of P. Sylvestris have anti– inflammatory, oxidation resisting and antivirus effects. Its major chemical constituents, including diterpenes, naphtha, flavones and so on, have been reported. Pharmacological experiments indicate that the P.E. and EtOAc extracts of this plant have significant antitumor and immunosuppressive activities. In order to search the bioactive consitituents from nature source, we began to study the chemical constituents from the needles of P. Sylvestris. By repeated column chromatography, including Silica gel, Toyopearl HW-40C and preparative HPLC, three new (1-3) and twenty-three compounds (4-26) were isolated from the EtOH extract of P. Sylvestris. By means of 1H-NMR,13C-NMR and 2D-NMR spectra, the structures of new compounds were elucidated as 15-ethyl-18-methyl pinifolate (1), 15-Acetoxy-labda-8(17),13E-dien- 18-al (2), 18-Hydroxy-labd-8(17),13E-dien-15-acetate (3). Compound 4 (15- Acetoxy-labd-8(17)-en-18-oic acid) is a new natural product, and seventeen compounds were isolated from P. Sylvestris for the first time as followed: 18α-Acetoxylabd-8(17)-en-15-oic acid (8), 3β-Hydroxy-8(17),E-13-labdadien-15- oic acid (10), Dehydroabietic acid (11), 7α-Hydroxydehydro-abietic acid(12), 7β-Hydroxydehydro-abietic acid (13), 7α,18-dihydroxydehydro-abietanol (14), pimarol (16), pimaric acid (17), isopimarol (18), isopimaric acid (19), 7-oxo-13-epi-pimara-8,15-dien-18-oic acid (20),α-Cadinol (21), T-Cadinol (22), eudesm-11-en-4α-ol (23), spathulenol (24), oplopanone (25) andβ-daucosterol (26).Our previous study showed that the P.E and EtOAc extracts revealed significant antitumor activities, and the further research for isolated compounds were screened by cytotoxity effects with Hela and A549 cells. Compound 12 revealed significant inhibitory effect on Hela and A549, compounds 7 and 13 showed positively inhibitory effect on Hela, and compounds 14 and 15 revealed positively inhibitory effect on A549.In the lymphocyte transformation experimention, the P.E. and EtOAc extracts revealed the significant inhibitory effect to the concanavalin (Con A) control group (p<0.01, n=6) and positively showed immunosuppressive activity. It is necessary to take a further research on the immunosuppressive constituents from P. Sylvestris.
