超声辐射微泡剂抑制Balb/c裸鼠SMMC-7721移植瘤的实验研究

【摘要】 目的建立SMMC-7721系肝癌Balb/c裸鼠移植瘤模型,研究低频超声辐射微泡剂对裸鼠SMMC-7721移植肿瘤体积增长抑制情况和肿瘤组织病理学变化,观察肿瘤局部SOD浓度,CD105表达情况和MVD的数量,研究肿瘤体积增长率和SOD,MVD之间的相关性。并初步探讨产生该结果的可能机制。方法调整SMMC-7721肝癌细胞浓度至5×106/ml,于每只裸小鼠左侧皮下注射0.2 ml建立SMMC-7721系肝癌Balb/c裸鼠移植瘤模型;模型建立成功后,50只裸鼠随机分为五组,对照组(A),单纯微泡组(B),单纯超声组(C),超声辐射微泡隔日组(D),超声辐射微泡每日组(E),每组10只模型裸鼠。各组采用相应的处理方法后,连续一周测量各组肿瘤体积大小,计算肿瘤体积增长率并绘制肿瘤体积增长曲线;取瘤体标本,石蜡切片后,HE染色电子显微镜下观察各组肿瘤组织病理学损伤;应用免疫组化的方法分析局部氧自由基代谢产物SOD浓度变化;并分析增长率与SOD浓度之间的相关性。同样用免疫组化法,观察各组肿瘤中CD105表达的差异,并由此对各组肿瘤微血管(MVD)进行计数,并分析MVD与肿瘤体积增长率之间的相关性。结果SMMC-7721肝癌细胞种植后第二周可于Balb/c裸小鼠皮下触及瘤结节。经相应处理一周后,各组间肿瘤体积大小各异(cm3):A组(0.207±0.081)、B组(0.216±0.019)、C组(0.180±0.015),A组肿瘤体积较D组(0.139±0.029)和E组(0.122±0.025)明显增大,(P<0.05);而A、B、C组间以及D、E组间则无统计差异。肿瘤体积增长率(%)的比较发现:D组(73.20±0.138),E组(57.24±0.1653)肿瘤体积增长率明显低于对照的A组(121.24±0.347)、B组(123.13±0.232)、C组(108.74±0.221),(P<0.01)。D和E组间无统计学差异,A、B和C组之间也无统计学意义。病理学检查见D,E组的瘤体标本内可见明显的血栓形成和大片梗死。免疫组化法得出局部SOD阳性率(%):D组(28.83±2.483)、E组(32.50±3.017)均高于A组(11.50±2.881),(P<0.01)而A、B、C组间以及D、E组间则无统计差异。并且SOD阳性率与肿瘤生长率间成负相关,(r=-0.663)。CD105免疫组化结果显示,A组、B组和C组的染色阳性较D组和E组明显增多,MVD计数结果显示:A组(64.6±2.03)、B(68.0±1.32)组和C(59.3±1.21)组比D(31.3±1.21)组和E(28.1±0.81)组显著增高,D,E两组与A组相比,具有统计学意义,(P<0.01)。A组、B组、C组间无统计差异,D组和E组间也无统计差异,MVD与肿瘤体积增长率之间成正相关(r=0.576)。

【Abstract】 Objective: To establish SMMC-7721transplantation tumor model in Balb/c athymic mouse. And investigate the Histopathology change and the restrained growth of SMMC-7721 hepatic cancer planted in athymic mouse after low frequency ultrasound radiate microbubble; the local concentration of local free-radicle and the expression of CD105 was observed, then explore the mechanism of this action.Methods: Adjust SMMC-7721 cell concentration to 5×106/ml, then subcutaneousely planted into balb/c mice for establishing models, 0.2ml evey mouse. After the model established ,fifty mice were randomly divided into five groups, control group(A), simplicity microbubble group (B), simplicity ultrasound(C), ultrasound and microbubble qd group (D) and ultrasound and microbubble qod group (E). After disposal correspondly, measure the volum of gross tumor for a week, Then, draw a growth curve and count the ratio of tumor growth, collect the tumor sample and histopathology damage was observed. The local concentration of local free-radicle was observed by immunohistochemistry with anti-SOD. relation between tumor growth ratio and concentration was analyzed.And observe the CD105 concentration in immunohistochemistry was observed with anti-CD105, then calculate the MVD of each group and analyz the relation between MVD and tumor growth ratio.Results: Two weeks later, the model established. After disposal correspondly for a week, tumor volume of each group has obviously difference(cm3) , the volume of group D(0.139±0.029) and group E(0.122±0.025) were significantly smaller than those of group A(0.207±0.081), (P<0.05). No statistics difference among groupA、B and C,so dose between group D and E. Statistical difference were been between group A(121.24±0.347)and group D(73.20±0.138) or E(57.24±0.1653)in ratio of volume growth(%), (P<0.01). Much thrombogenesis and infarctus can be observed in histopathology in group D and group E Concentration of SOD (%)in tumor tissue of group D (28.83±2.483)and E(32.50±3.017)
