镁对高脂高糖膳食诱导大鼠肥胖的影响及机制

【摘要】 随着肥胖人口及肥胖相关疾病发病率逐年增加,肥胖的饮食防治已成为营养学领域的研究热点。目前,矿物质和维生素与肥胖的关系越来越引人注目。肥胖与人体能量代谢异常密切相关,镁(Magnesium,Mg)是人体必需的常量元素之一,催化和激活体内300多种酶系,参与体内所有能量代谢过程,并在能量的运输、贮存及利用过程中发挥关键作用。虽然目前有关镁与超重和肥胖的研究较少,但最近的研究发现,肥胖者处于负镁平衡状态,膳食镁摄入低于体重正常者;肥胖者血清镁、红细胞镁和血小板镁均低于体重正常者,血清镁与实际体重/理想体重呈负相关;各种治疗肥胖的措施,如禁食、药物、外科手术等都会造成机体镁的丢失。镁可能是肥胖、糖尿病、动脉粥样硬化等肥胖相关疾病的主要危险因子。另有人群流行病学调查得出摄入充足的镁有助于预防肥胖及由此引起的心血管疾病。本研究配制半合成低镁基础饲料,建立高脂高糖饲料诱导大鼠肥胖模型的同时补镁,观察镁对肥胖的影响,并探讨其机制,探索肥胖饮食疗法的新途径,同时深入研究镁的营养保健作用,为镁的合理营养提供理论依据。 1.补镁对高脂高糖膳食诱导大鼠肥胖的影响 本研究在高脂高糖膳食诱导大鼠肥胖的同时补充镁,观察大鼠模型的代谢特点和病理变化,探讨补镁对高脂高糖饲料诱导大鼠肥胖进程的影响。实验分组为:①普通饲料组;②低镁饲料组;③高脂高糖饲料组;④补镁组:高脂高糖饲料+500mg/kg氧化镁、高脂高糖饲料+1000mg/kg氧化镁、高脂高糖饲料+2000mg/kg氧化镁。各组实验动物喂以相应的饲料,共9周。实验结束时处死动物,检测体重、脂/体、脏器系数、血清和骨骼肌镁含量、血清总胆固醇(total cholesterol,TC)、甘油三脂(triglyceride,TG)、高密度脂蛋白(high-density lipoprotein,HDL)、极低密度脂蛋白(very low-density lipoprotein,VLDL)、空腹血糖、胰岛素,并计算胰岛素抵抗指数(homeostasis model assessment-insulin resistance index,HOMA-IR)和胰岛素敏感指数(insulin resistance index,ISI)。

【Abstract】 Obesity and obesity related diseases are increasing in prevalence. Obesity can only occur when the energy value of food eaten exceeds energy expended. Magnesium (Mg) is one of the necessary macroelement in the human body, and is involved in more than 300 different enzymatic reactions, makes effect on almost all energy metabolisms. People who have obesity or obesity related diseases may have increased Mg utilization or Mg deficiency. Significant insights into the etiology of human obesity have been gained from the study of animal models. The present experiment was planned to study the effect of Mg supplementation on high fat and sugar diet induced obese rats and its probable mechanism. 1 Development of obesity animal modelsMg was supplemented into the high fat and sucrose diet during the experiment. Metabolism and pathology changes were observed to study the effect of Mg supplementation on high fat and sucrose diet induced obese rats. The rats were divided into six groups: the normal diet group, the low Mg diet group, high fat and sucrose diet group and Mg supplementation groups (500mg/kgMgO, 1000mg/kgMgO, 2000mg/kgMgO). The animals were killed after 9 weeks, the body weight, VF/weight, organ coefficient, blood and muscle Mg content and other obesity parameters were measured. Abdominal fat tissue and skeletal muscle were collected to be made in paraffin-embeded specimens.The results showed that the weight and weight gain of high fat and sucrose diet group were higher significantly than the low Mg diet group (P<0.05). The VF/weight and liver coefficient of high fat and sucrose diet group was higher significantly than the low Mg diet group (P<0.05, P<0.01). The level of serum TC, TG, VLDL of high fat and sucrose diet group was higher significantly than the low
