胰岛素样因子Ⅰ、血管生成素Ⅱ和色素上皮源性生长因子在低压性缺氧成年大鼠视网膜的表达

【摘要】 目的:探讨成年大鼠在低压性缺氧后,胰岛素样因子I,血管生成素Ⅱ和色素上皮源性生长因子在其视网膜的表达情况。方法:成年Wistar大鼠暴露于低氧环境(9-7%O2)2h后,分别在3h、24h、3d、7d和14d,通过实时荧光定量逆转录聚合酶链反应,蛋白免疫印迹法和免疫组化的方法检测胰岛素样因子I(IGF-I),血管生成素Ⅱ(AngⅡ)和色素上皮源性生长因子(PEDF)在视网膜的表达情况。结果:在暴露于低氧性环境后的3h,24h和3d,成年大鼠视网膜IGF-I,AngⅡ mRNA和蛋白质水平均较正常对照组增加(P<0.05),在7d和14d其表达较正常对照组差异无显著性。PEDF mRNA和蛋白质水平在缺氧后3h、24h、3d、7d和14d均明显低于正常对照组(P<0.05)。结论:这次实验结果表明缺氧可以使成年大鼠视网膜IGF-I、AngⅡ表达上调,PEDF的表达下调。这种促血管生成因子和抑制血管生成因子的差异性表达可能会共同导致了视网膜新生血管的形成,导致视网膜的损伤。
【Abstract】 Objective: The present study was undertaken to examine the expression of Insulin-like growth factor I(IGF-I), Angiopoietin Ⅱ(Ang Ⅱ)and Pigment epithelium-derived factor(PEDF) in the retina of adult rat following hypobaric hypoxic exposures. Methods: Adult Wistar rats were subjected to hypobaric hypoxia for 2h following which expression of IGF-I, Ang Ⅱ and PEDF in the retina was investigated from 3h-14d by real time RT-PCR, western blotting and immunohistochemistry. Results: Upregulated mRNA and protein expression of IGF-I, Ang II in the retina was observed at 3h, 24h and 3d after hypoxic exposure following which it decreased at 7d and 14day. PEDFmRNA and protein expression in the retina was decreased at 3h-14d following exposure to hypoxia. Conclusions:The findings of this study indicate hypoxia can upregulate IGF-I, Ang Ⅱ and downregulate PEDF in adult retina. The differential expression of pro- and antiangiogenic factors of IGF-I, Ang Ⅱ and PEDF may be involved in hypoxic damage to the retina. These vasoactive factors together may be responsible for retinal neovascularisation.
