白芍双苷注射液抗肿瘤活性的研究

【摘要】 本文从白芍双苷注射液的抗肿瘤活性入手,研究了其对免疫功能抑制的改善作用以及其的升白作用。 建立小鼠移植性骨肉瘤S180和MathA肿瘤实验动物模型,观察白芍双苷注射液的抗肿瘤作用。实验结果表明:在所选择的剂量范围内(12.5mg/Kg、6.25mg/Kg、3.125mg/Kg),白芍双苷注射液显著抑制小鼠肉瘤180(S180),抑制率分别为55.5%、47.2%和39.6%;显著延长荷MathA腹水型肿瘤小鼠的生存时间,生命延长率分别为78.4%、44.6%和31.7%;并且具有显著的免疫调节作用:恢复荷瘤小鼠巨噬细胞吞噬功能,上调脾巨噬细胞NO、腹腔巨噬细胞TNF-α、脾细胞IFN-γ的分泌和增强NK细胞的杀伤活性以及促进脾淋巴细胞增殖。 通过制备免疫低功小鼠实验动物模型,观察白芍双苷注射液对免疫功能抑制的改善作用,实验结果表明:在所选择的剂量范围内(12.5mg/Kg、6.25mg/Kg、3.125mg/Kg),白芍双苷注射液对免疫功能低下小鼠具有显著的改善作用,恢复环磷酰胺所致免疫低功模型小鼠巨噬细胞吞噬功能、增强NK细胞的杀伤活性,促进脾淋巴细胞增殖,升高外周血白细胞总数并使脾脏指数上升。 通过制备小鼠和家兔白细胞减少实验动物模型,观察白芍双苷注射液的升白作用,实验结果表明:在所选择的剂量范围内,其中12.5mg/Kg、6.25mg/Kg、3.125mg/Kg剂量的白芍双苷注射液不仅可以提升X-射线照射和环磷酰胺致小鼠的外周血白细胞总数、血小板数量、血红蛋白含量和骨髓有核细胞总数,还可使外周血及骨髓白细胞分类趋于正常,减轻X-射线照射和环磷酰胺注射所造成的骨髓抑制作用,并使脾脏指数升高;4.64mg/Kg、2.32mg/Kg、1.16mg/Kg剂量白芍双苷注射液可以提升盐酸阿糖胞苷致家兔的外周血白细胞、血小板数量和血红蛋白含量,并使外周血及骨髓白细胞分类趋于正常,减轻盐酸阿糖胞苷所造成的骨髓抑制,并使脾脏指数升高。 综合以上实验结果提示:白芍双苷注射液具有一定的抗肿瘤活性,其作用机制可能通过调节机体免疫系统而发挥抗肿瘤作用;对环磷酰胺所致的小鼠免疫功能低下具有一定的改善作用;对X-射线照射、环磷酰胺、盐酸阿糖胞苷导致的受试动物白细胞减少及骨髓抑制具有明显的改善作用,以上作用均呈现一定剂量依从。

【Abstract】 Antitumor actived, immunological enhancing effects and leukogenic effects of Baishao shuanggan injection were investigated in this paper.Antitumor activites were assayed by using mice carring transplanted sarcoma S180 and MethA as an animal model. Baishao shuanggan injection at dosage of 12.5, 6.25 , 3.125mg·kg-1 has obvious inhibit effect for mice bearing S180 tumor, the inhibitition rate of 55.5%, 47.2%, 39.6% and obvious prolonged the survival time of mice bearing MethA tumor, the life prolong rate of 78.4%, 44.6%, 31.7%. At the dose as above has obvious immunomodulating effect for mice bearing S180 tumor. The phagocytosis function of celiac macrophage could be recovered, the activity of NO from the spleen cells was improved, the production of TNF-α from the supernatant of peritoneal macrophages and IFN-γ from spleen cells was enhanced , the cytotoxicity activities of NK from the spleen cells was enhanced, furthermore the T lymphocytes proliferation could be increased.Immunological enhancing effects were studied by using immunosuppressed mice as an animal model. Baishao shuanggan injection at dosage of 12.5,6.25,3.125mg·kg-1 has obvious enhanced immunological for immunosuppressed mice. The phagocytosis function of celiac macrophage could be recovered, the cytotoxicity activities of NK from the spleen cells was enhanced, the white blood cell count of peripheral blood was enhanced, and the coefficients of spleen was improved.Leukogenic effects were studied on mice or rabbit leukopenia models. Baishao shuanggan injection at dosage of 12.5, 6.25, 3.125mg·kg-1 could obviously increase the counts of white blood cell, platelet, bone morrow karyocyte and the content of hemoglobin, could come into being normal about the classify of peripheral blood and bone marrow, could ease arrest of bone marrow , furthermore could improve the coefficients of spleen on mice of raying by X-ray and inject by cyclophosphamide. Baishao shuanggan injection at dosage of 4.64, 2.32, 1.16 mg·kg-1 could obviously increase the counts of white blood cell, platelet, bone morrow karyocyte and the content
