半枝莲化学成分研究

A Study on the Chemical Consituents of Scutellaria Barbata D. Don

【摘要】 半枝莲(Scutellaria barbata D.Don)为唇形科黄芩属植物,别名狄叶韩信草、并头草、牙刷草等,以干燥全草入药。其名最早见于《外科正宗》,味辛、苦,性寒,归肺、肝、肾经,具有清热解毒、散瘀止血、利尿消肿的功效,用于疔疮肿毒、咽喉肿痛、毒蛇咬伤、跌扑肿痛及水肿、黄疸。现代药理学研究表明,半枝莲的水提物和醇提物均有明显的抗肺癌、消化系统癌、肝癌、乳腺癌、绒膜上皮癌的活性。笔者采用MTT法对半枝莲70%乙醇提取物的正丁醇萃取物和氯仿萃取物进行了体外癌细胞生长抑制活性测试,发现:正丁醇层萃取物对Sf188细胞和H460细胞均表现出明显的生长抑制活性,对Sf188细胞更为显著;氯仿层萃取物对H460细胞表现出较为明显的生长抑制活性,而对Sf188细胞的抑制不明显。为寻找半枝莲中具有抗癌活性的有效成分,对半枝莲氯仿萃取物和正丁醇萃取物的化学成分进行了研究。从其70%乙醇提取物中分离得到14个化合物,结合理化性质及核磁共振谱学分析,并与文献对照,分别鉴定为:对羟基苯甲醛(1)、3,4-二羟基苯甲酸(2)、对羟基苯乙李玉酮(3)、5,7,4’-三羟基-8-甲氧基黄酮(4)、5,7,4’-三羟基-6-甲氧基黄酮(5)、5,7,4’-三羟基黄酮(6)、木樨草素(7)、5,4‘-二羟基-6,7,3’,5’-四甲氧基黄酮(8)、5,7,4’-三羟基二氢黄酮(9)、5-羟基-4’-甲氧基黄酮-7-O-α-L-鼠李糖基-(1→6)-β-D-吡喃葡萄糖苷(10)、半枝莲酸(11)、香豆酸乙酯(12)、β-谷甾醇(13)、胡萝卜苷(14)。其中化合物8,10,12为该植物中首次分离得到。

【Abstract】 Scutellaria barbata (D. Don), labiatae, is a traditional Chinese medicine, and widely used as an anti-inflammatory and a diuretic drug. The whole plant can be used for medicine. Several studies have indicated that extracts of S. barbata have growth inhibitory effects on a number of human cancers, and its treatment uses for lung cancer, digestive system cancers, hepatoma, breast cancer, and chorioepithelioma have been reported for a long time.From the 70% ethanol extract of the whole herb of S. barbata, by solvent extraction, repeated column chromatography and recrystallization, 14 constituents were isolated. On the basis of physico-chemical properties and spectroscopic analysis, they were identified as follows: p-hydroxybenzaldehyde (1), 3,4-dihydroxybenzoicacid (2), p-hydroxyaceophenone (3), 5,7,4’-trihydroxy-8-methoxyflavone (4), 5,7,4’-trihydroxy-6-methoxyflavone (5), 5,7,4’-tr -ihydroxyflavone (6), luteolin (7), 5,4’-dihydroxy-6,7,3’,5’-tetramethoxyflavone (8), naringenin (9), 5-hydroxy- 4-methoxyflavone-7-O-α-L-rhamnosyl-(1→6)-β-D-glucopyranoside (10), scutellaric acid (11), p-coumaric acid acetate (12), β-sitosterol (13), daucosterol (14). Among them, compounds 8,10,12 were isolated from the plant for the first time.The MTT experiments in vitro showed that the chloroform and n-butanol extracts of S. barbata had significant growth inhibitory effects on the H460 and Sf188 cancer cells.
