食用菌液体培养参数优化及接种装置研究

【摘要】 本课题针对目前我国食用菌生产企业大都使用固体菌种生产,产业化水平较低的现状,以将食用菌液体菌种生产技术从实验室走向生产企业为目标,选择食用菌珍稀品种鸡腿蘑为研究对象,研究了鸡腿蘑液体菌种培养的营养需求和适宜的环境条件,并在发酵罐中进行扩大培养,建立鸡腿蘑菌丝的生长模型,为工厂化大规模生产提供技术支持。同时,为使液体菌种能顺利应用于工厂化生产中,针对目前接种技术及设备不过关的现状,研制了液体菌种培养接种一体化技术及装置。 主要研究结果如下: (1)从五种不同的鸡腿蘑菌株(白鸡、北鸡、兴野、防鸡、RJ)中,筛选出菌球小而多、生物量高的菌株RJ作为液体培养的菌株。测定鸡腿蘑RJ生长过程中菌丝干重及还原糖的含量,根据菌丝干重和还原糖的变化规律,确定了鸡腿蘑液体培养的最佳时间为5天。 (2)以鸡腿蘑RJ为研究对象,通过单因素试验和正交试验,确定了最佳液体培养基配方为:葡萄糖2%,玉米粉2.5%,酵母浸出汁0.5%,麸皮4%,KH2PO4 0.25%,MgSO4 0.1%,添加0.05mg/L的VB1和0.5%的豆油。 (3)通过单因素试验和正交试验优化了鸡腿蘑RJ液体培养的最适温度、pH值和溶解氧。最适温度为24℃、最适初始pH值为6.5、三角瓶装液量为400ml/500ml。用10L发酵罐进行扩大培养,与三角瓶培养相比,鸡腿蘑RJ菌丝生长的延迟期缩短了12小时,最佳培养时间缩短了36小时,菌丝干重的最大值也提高了41.28%。 (4)针对不同液体菌种菌丝大小不同,接种量不同,需要采用插入接种或者表面布种方式等特点,利用液体容积式定量原理,采用电磁阀控制,研制出一套自动化程度高、接种量可以调节、接种嘴可以更换、适应不同液体菌种要求的、染菌率低的制种与接种一体化装置。

【Abstract】 At present, most of the edible fungi factories in China are still using solid spawns in cultivation, but they have not been industrialized. In order to apply liquid spawns into edible fungi factories, the nutrition demands and environment conditions of Coprinus comatus (Mull.:Fr.) Gray were studied and an integrative device of edible fungi liquid spawns cultivation and inoculation was designed in this thesis.The main conclusions are as follows:(1) It turned out that the mushroom of RJ had the smallest mycelium and the most the highest mycelial yield. So the mushroom of RJ was the best among five different varieties of baiji, beiji, xingye, fangji, RJ for liquid culture.The mycelial yield and deoxidize sugar were measured during the culture process. It showed that the best time for liquid culture of RJ was five days.(2) The best medium of RJ was known from the single-factor and orthogonal experiments, results showed that the best medium is as follows glucose 2%, corn flour 2.5%, yeast extract 0.5%, bran 3%, KH2PO4 0.25%, MgSO40.1%, VB1 0.05mg/L and bean oil 0.5%.(3) The suitable temperature, pH, DO were studied through the single-factor and orthogonal experiments, the results showed that the most suitable condition were the temperature of 24°C、pH 6.5、 and with 400ml medium in 500ml flask. When culturing Liquid spawns in ten liters, the lag phase was shortened by twelve hours, the best culture time was shortened by thirty-six hours and the mycelial yield was increased by 41.28%, compared with culturing them in flasks.(4) An integrative device of edible fungi liquid spawns cultivation and inoculation was designed, which realized the automation of inoculation, the adjustment of inoculum and the exchange of the mouth of inoculation.
