万寿菊雄性不育RAPD分子标记的研究

【摘要】 万寿菊是现代园林中广泛应用的草本花卉,也是极具利用价值的经济植物。本研究找到了快速提取万寿菊雄性不育系、可育系、保持系和杂种F1代DNA的方法,采用了3次正交试验设计建立了RAPD反应扩增体系。然后,运用该扩增体系在五组随机引物中筛选出了引物OPA18进行万寿菊种子纯度的鉴定,得到了与田间检测接近的种子纯度结果。最后,选择万寿菊核不育两用系AALY001为材料,对雄性不育株和可育株进行扩增,得到了一条在不育株中缺失,在可育株中出现的稳定的特异标记NMSOPM09500。因此,用RAPD分子标记技术能够较有效地进行万寿菊种子纯度的鉴定,并且也能够为雄性不育万寿菊不育基因的研究提供一定的技术支持。
【Abstract】 Marigold is the popular flower used in the modern landscaping and also the valuable economic plant. The research developed a quick method to extract DNA from marigold male sterile line, fertile line, maintainer line and F1. And a RAPD reaction system suitable for marigold was developed by thrice orthogonal experiment designs. Secondly, the results showed that the primer OPA18 can be considered as the marker to identify the seed purity of marigold stably.The identification data of seeds purity by RAPD technique using primer OPA18 almost accorded with the regular identification data of it in the field. Thirdly, using RAPD technique to amplify DNA of the AALY001 NMS two lines, one specific marker NMSOPM09500 was obtained in the male fertile line but without in the male sterile line. This marker was stable after repeating amplification. Therefore, it is helpful to identify seed purity and it also provides technical support for the research on male sterile gene of male sterile marigold by RAPD marker.
