桑黄多糖提取、分离纯化及理化性质研究

【摘要】 本论文对桑黄多糖的提取、分离纯化、理化性质及菌丝体深层发酵进行了研究,建立了桑黄多糖分离纯化的技术路线,并对不同来源桑黄的粗多糖含量和蛋白质含量等进行了比较。 将桑黄多糖依次采用乙醇分级醇沉、超滤、离子交换层析和凝胶过滤等分离纯化方法得到一个均一组分PBF1-A。分级醇沉将桑黄多糖初步分为两个部分PBF1和PBF2,得率分别为43%和49%;然后对PBF1进行超滤,多糖纯度达到76%,回收率为86%;将超滤后的滞留液采用离子交换层析,然后脱盐、浓缩,再经凝胶过滤得到均一组分PBF1-A。 桑黄多糖均一组分PBF1-A极易溶于水,是一种蛋白糖,其多糖含量为96.6(±0.5)%,蛋白质含量为3.04(±0.05)%,比旋度为-4.6°,分子量大于2000KDa;用三氟乙酸对PBF1-A进行完全酸水解,可知PBF1-A的单糖残基由葡萄糖、甘露糖、木糖、半乳糖和岩藻糖组成,摩尔比分别占75.09%、9.11%、7.32%、4.39%和4.10%;红外光谱和核磁共振谱表明PBF1-A主要含有β-吡喃葡聚糖,此外含有甘露糖,与酸水解结果一致。 采用液体深层发酵生产桑黄菌丝体,确定培养基组成为:玉米粉
【Abstract】 The extraction, purification, and physicochemical properties of water-soluble polysaccharide fractions from the fruiting body of Phellinus baumii Pilat were studied in this paper. Then the process flow of purification of polysaccharide was established. The submerged fermentation of Phellinus baumii Pilat was also preliminary studied. The content of crude polysaccharide and crude protein et al from various Phellinus baumii Pilat were compared.A homogeneous fraction designated PBF1-A from Phellinus baumii Pilat was isolated by gradient ethanol precipitation, ultrafiltration, ion exchange chromatography and gel chromatography. Two parts, designated PBFl and PBF2 which yield was 43% and 49% respectively, were isolated from crude polysaccharide by gradient ethanol precipitation. The PBF1 was
