丁香酚引起小鼠空间学习记忆行为学改变及其机制

【摘要】 研究背景及研究目的 神经系统疾病药物治疗过程中,由于BBB的特殊结构,药物或者其它的外源性化学物质要想进入CNS,依赖于BBB和血脑脊液屏障,以及这些物质的理化性质。嗅觉系统中提供了大脑与外部环境的直接联系,嗅觉的脑通路可能成为CNS药物直接进入脑的一条新的途径。丁香酚作为芳香开窍类药物,其醒脑开窍的机制是什么,一直未能深入研究,本研究试图探讨丁香酚通过嗅觉途径对动物空间学习记忆影响的机制。 第一部分 丁香酚不同途径给药对昆明鼠的神经行为学观察 比较丁香酚不同给药途径对昆明鼠学习记忆等神经行为学的影响,选择了嗅觉吸入组,鼻腔滴入组,灌胃组不同的给药途径,应用神经行为学的方法,观测小鼠自主活动及空间学习记忆能力的改变。 结果 与鼻腔滴入组,灌胃组和空白组比较,丁香酚嗅觉吸入组能够明显改善小鼠的空间学习记忆。灌胃组能够明显增强小鼠的自主活动能力。 第二部分 空间学习记忆障碍小鼠以及嗅球摘除后小鼠水迷宫实验观察 进一步使用学习记忆障碍小鼠和制作嗅球摘除模型给予嗅觉吸入丁香酚后的Lashley Ⅲ水迷宫结果,验证丁香酚是否具有增强学习记忆的能力及学习记忆是否与嗅觉之间存在关联。通过吸入丁香酚观察,采用Lashley Ⅲ水迷宫,探讨丁香酚改善小鼠空间学习记忆能力是否与通过嗅觉途径有关。 结果 假手术组、模型组的自主活动高于空白组小鼠自主活动。学习记忆障碍药物组水迷宫结果与学习记忆障碍空白组比较,有显著性差异。嗅球摘除组以及摘除

【Abstract】 Backgrounds and ObjectivesBecause of the special structure of BBB, medicine and other xenobiotics can enter CNS depending on the presence of BBB and Blood-CSF as well as the physico-chemical properties. As the Olfactory system provides a direct pathway through inner brain to outer, the olfactory-to-brain pathway may be a new way for the CNS medicines’ entry into the inner brain. Eugenol is one of the fragrance enlightened medicines,but its mechanisms of englightening has not been well studied.This research tries to approach the mechanisms of eugenol’s influences on space learning and memory of animals though olfactory pathway.If Eugenol ,one of the fragrance enlightened medicine, can be administered through olfactory inspiratory way, then this pathway can be admitted as one of the most effective and simplest way for medicines’ entry into CNS.This paper is to explore the mechanism of influences on learning and memory after Eugenol’s administration through olfactory pathway. This paper can be divided into three parts:The first part: The neuroethological observations on Kunming mice after Eugenol’s administration through different pathways.Methods To compare the different influences on Kunming mice after Eugenol’s administration through different ways, three groups were slelected: Olfactory inhaling group; Nasal gutted group and Intragastric administration group. The changes of mice autonomic activities and learning and memory can be observed through neuroethology methods.
