高γ-氨基丁酸、低咖啡碱绿茶急性毒性和辅助降血压功能评价

【摘要】 γ-氨基丁酸又名氨酪酸(非蛋白质氨基酸),是哺乳动物中枢神经系统最主要的抑制性氨基酸神经递质,它参与多种代谢活动,具有很高的生理活性。20世纪80年代日本的津志先生首先采用工程方法提高绿茶中γ-氨基丁酸含量,用于保健茶制造。本实验研究比较了用不同茶样的不同浸提方法浸提绿茶样,并测定茶汤中γ-氨基丁酸、咖啡碱和其它常规成分含量,以γ-氨基丁酸含量较高、咖啡碱含量较低、其它成分含量相对较高为选择依据,比较选用福选9号(F9)、福云6号(F6)茶鲜叶在绿茶正常加工工艺的基础上再经过15h嫌气和脱咖啡碱处理的茶样F915h水潦和F615h水潦为急性毒性试验样品,选用F615h水潦、福云6号经过15h嫌气处理的F615hs和福云6号不经嫌气处理的普通绿茶F6X0Sn为降血压功能评价样品。参照我国保健食品降血压功能评价方法和急性毒性实验方法,以原发性高血压大白鼠(SHR)和清洁级昆明种小白鼠为实验对象,对高γ-氨基丁酸、低咖啡碱绿茶的辅助降血压功能和急性毒性进行了评价,并对其降压机理进行初步探讨。以此推动无副作用的纯天然保健降压茶的开发与研究。实验结果如下: 1.每天一次性大剂量经口给小白鼠灌胃受试茶样,经过两周实验观察,各组动物饮水和摄食量正常,神经精神症状及活动情况正常,未出现动物死亡状况。不能得出具体的LD50(mg/(kg.bw)),但根据最大灌胃剂量,可以判断两个受试茶样的LD50>5000mg/(kg.bw),依据化学物质急性毒性分级标准确认两茶样均属于实际无毒物质。 2.经过40d的辅助降血压功能评价试验后得出,用中剂量(0.42g/(kg.bw))的F615h水潦、F615hs和高剂量(1.05g/(kg.bw))的F615h水潦和中剂量(0.8mg/(kg.bw))GABA标准溶液样品每天灌胃大鼠,在试验结束时,大鼠的收缩压较初始值有显著降低;且F615h水潦高剂量组的降压效果达到极显著水平。用同等中剂量的F6X0Sn和低剂量的(0.21g/(kg.bw))F615h水潦样品灌胃后大鼠收缩压也有一定的降低,但无统计学意义;空白对照组大鼠收缩压有一定的上升,没有统计学意义。在相同中等灌胃剂量下,样品F615h水潦的降压效果明显好于F615hs;说明脱去咖啡碱可以辅助提高GABA的降压效果;用相同GABA含量的标样灌胃后的降压效果也没有F615h水潦好,说明除了GABA外,茶样中的其他生理活性成分也有一定

【Abstract】 After comparing different lixiviation methods of green tea ,using time after time and multiplestep lixiviation method to lixiviate tea samples,then determined GAB A、 caffein and other general ingredients in tea soup.Constrasting test results and according to the principle of high GABA、 lower caffeine and other ingredients relatively high,chose F915h 水潦和 F615h 水潦 (Fuxuan 9 and Fuyun 6 .dealt with 15h absence of oxygen and left off caffein ) as acute toxicity evaluation samples , F615h 水潦、 F615hs (Fuyun 6 dealt with 15h absence of oxygen) and F6XoSn(general green tea) as assisting blood pressure reduction function evaluation samples.Consult assisting blood pressure reduction fuction and acute toxicity evaluation standard in our country ,the study used spontaneously hypertensive rat and white mice (KM) as test animals to evaluation acute toxicity and assisting blood pressure reduction function of these tea samples ,also primarily discuss the blood pressure reduction mechanism.The study can ensure these teas put into market safely、 upgrade tea value、 drive to tap of natural and little side-effect health food. The results is as followings:1. Pouring larger dose tea soup one-time into white mice from mouth to stomach every day, every group mice drinking water 、 ingestion、 nerve and activity were all normal after two weeks and no mice was died.It could not draw definite LD50(mg/(kg.bw)) ,but according to the largest pouring dose ,we could judge that LD50>5000mg/ (kg.bw) of the two tea samples,according to chemical substance acute toxicity classification standard,we can say that these tea samples are all innocuity.2. Pouring 0.42g/ (kg.bw) F615h 水潦、 F615hs,1.05g/(kg.bw)F615h 水潦 and 0.8mg/ (kg.bw) GABA standard solution into SHR stomach every day ,40ds later, systolic pressure of SHR reduced obviously compared to initial value and 1.05g/(kg.bw) F615h 水潦 had great effect. 0.42g/(kg.bw) F6XoSn and 0.21g/(kg.bw) F615h 水潦 also could reduce blood pressure ,but it did not have
